目的,2,基于PI3K/AKT通路研究黄金胶囊干预2型糖尿病(T2DM)大鼠胰岛素抵抗的初步机制。,方法,2,选取SPF级Wistar雄性大鼠40只,随机选取其中的10只作为空白对照组,其余30只大鼠建立T2DM模型。建模成功后随机分为3组,每组10只,分别设为模型组、二甲双胍组、黄金胶囊组。二甲双胍组给予盐酸二甲双胍干预(0.2 g/kg),黄金胶囊组给予黄金胶囊干预(1.265 g/kg),模型组及对照组给予等容积0.9%NaCl溶液灌胃;1次/d,连续干预10周。测定各组大鼠血糖水平[空腹血糖(FPG)、糖化血红蛋白(HbAlc)]、PI3K/AKT信号通路蛋白[磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)、葡萄糖转运蛋白4(GLUT-4)]及胰岛素抵抗指数(HOMA-IR)]表达水平。,结果,2,与空白对照组大鼠比较,模型组大鼠FPG、HbAlc、FINS、HOMA-IR水平升高,PI3K、AKT、GLUT-4表达降低,差异有统计学意义(,P,<,0.05)。与模型组比较,二甲双胍组、黄金胶囊组大鼠FPG、HbAlc、FINS、HOMA-IR水平降低,PI3K、AKT、GLUT-4表达升高,差异有统计学意义(,P,<,0.05)。,结论,2,黄金胶囊可提高T2DM大鼠PI3K、AKT、GLUT-4表达,其可能通过激活PI3K/AKT信号通路降低大鼠血糖水平,从而改善胰岛素抵抗。
Objective,2,To explore the preliminary mechanism of intervention with gold capsule on insulin resistance (IR) in rats with type 2 diabetes mellitus (T2DM) based on the PI3K/AKT pathway.,Methods,2,Forty SPF-grade Wistar male rats were selected, 10 of which were randomly chosen into a blank control group, and a T2DM model was established in the remaining 30 rats. Those successfully modeled rats were assigned to model, metformin and gold capsule groups randomly, with 10 rats in each group. The metformin group was administered with metformin hydrochloride (0.2 g/kg), the gold capsule group was administered with gold capsule (1.265 g/kg) and the model group and control group were administered with equal volume of 0.9% NaCl solution by gavage once a day for 10 consecutive weeks. The blood glucose levels [fasting plasma glucose (FPG), glycated hemoglobin (HbAlc)], PI3K/AKT signaling pathway protein expression levels [PI3K, AKT, glucose transporter protein 4 (GLUT-4)] and homeostatic model assessment for insulin resistance (HOMA-IR) level were tested in each group.,Results,2,Compared with the rats in the blank control group, the rats in the model group had higher levels of FPG, HbAlc, FINS and HOMA-IR, but lower expression levels of PI3K, AKT and GLUT-4, and the differences were statistically significant (,P,<,0.05). Compared with the rats in the model group, the rats in the metformin and gold capsule groups showed lower levels of FPG, HbAlc, FINS and HOMA-IR, but higher expression levels of PI3K, AKT and GLUT-4, and the differences were statistically significant (,P,<,0.05).,Conclusion,2,Gold capsule can increase the expression of PI3K, AKT and GLUT-4 in T2DM rats, which may work by activating PI3K/AKT signaling pathway to reduce blood glucose levels and improve IR in rats.
