目的,2,观察不同剂量知葛通脉颗粒对糖尿病动脉粥样硬化(AS)大鼠斑块组织肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)、血管细胞黏附分子-1(VCAM-1)、细胞间黏附分子-1(ICAM-1)、基质金属蛋白酶组织抑制因子(TIMP)的蛋白及mRNA表达水平的影响。,方法,2,将70只4周龄SPF级Wistar雄性大鼠,随机选出10只为正常组,只给予基础饲料,其余60只为糖尿病造模组。造模组大鼠采用30 mg/kg剂量链脲佐菌素(STZ)注射+主动脉内膜球囊拉伤术加高脂饲料喂养制作糖尿病动脉粥样硬化大鼠模型。造模成功大鼠按血糖水平随机分为模型组、知葛通脉颗粒低剂量组(简称低剂量组,灌胃给予知葛通脉颗粒1.05 g·kg,-1,·d,-1,)、知葛通脉颗粒中剂量组(简称中剂量组,灌胃给予知葛通脉颗粒2.1 g·kg,-1,·d,-1,)、知葛通脉颗粒高剂量组(简称高剂量组,灌胃给予知葛通脉颗粒4.2 g·kg,-1,·d,-1,)及瑞舒伐他汀钠组(简称西药组,灌胃给予瑞舒伐他汀钠1.05 mg·kg,-1,·d,-1,),每组10只。给药6周后腹主动脉剪取粥样斑块留作标本,检测TNF-α、MMP-9、VCAM-1、ICAM-1、TIMP 的蛋白及mRNA表达水平。,结果,2,与模型组比较,高剂量组、西药组均能明显降低TNF-α、MMP-9、VCAM-1、ICAM-1、TIMP蛋白及mRNA表达水平,差异有统计学意义(,P,<,0.05);与模型组比较,低剂量组、中剂量组,TNF,-,α, mRNA表达降低,MMP-9、VCAM-1、ICAM-1、TIMP 的蛋白及mRNA表达水平降低,差异有统计学意义(,P,<,0.05)。与低剂量组比较,高剂量组能明显降低TNF-α、MMP-9、VCAM-1、MCP、ICAM-1、TIMP蛋白表达水平,降低,TIMP, mRNA表达水平,差异有统计学意义(,P,<,0.05);与低剂量组比较,中剂量组能明显降低VCAM-1、TIMP蛋白表达水平,差异有统计学意义(,P,<,0.05)。与中剂量组比较,高剂量组能明显降低MMP-9、ICAM-1、TIMP蛋白表达水平,差异有统计学意义(,P,<,0.05)。,结论,2,知葛通脉颗粒能明显降低糖尿病AS大鼠斑块组织TNF-α、MMP-9、VCAM-1、ICAM-1、TIMP蛋白及mRNA表达水平,降低炎症因子的表达,改善血管内皮的炎症反应,高剂量组疗效优于中、低剂量组。
Objective,2,To observe the effects of different doses of Zhige Tongmai Granules (ZGTMG) on the expression of protein and mRNA of tumor necrosis factor-α (TNF-α), matrix metalloproteinase-9 (MMP-9), vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule-1 (ICAM-1) and tissue inhibitor of matrix metalloproteinase (TIMP) in plaque tissues of diabetic atherosclerosis (AS) rats.,Methods,2,Ten of 70 SPF-grade Wistar male rats at 4 weeks of age were randomly selected as the normal group and administered with basic feeds only, and the remaining 60 were the diabetic modeling group. The diabetic atherosclerotic rat model was designed by streptozotocin (STZ) injection (30 mg/kg), intra-aortic balloon pump injuries and the high-fat diet. The modeled rats were randomly divided into a model group, ZGTMG low-dose group (low-dose group, gavage administration of ZGTMG at 1.05 g·kg,-1,·d,-1,), ZGTMG medium-dose group (medium-dose group, gavage administration of ZGTMG at 2.1 g·kg,-1,·d,-1,), ZGTMG high-dose group (high-dose group, gavage administration of ZGTMG at 4.2 g·kg,-1,·d,-1,) and rosuvastatin sodium group (modern medicine group, gavage administration of rosuvastatin sodium at 1.05 mg·kg,-1,·d,-1,) according to the blood glucose level, with 10 animals in each group. After 6 weeks of administration, atheromatous plaques were obtained from the abdominal aorta as the specimen and the protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP were detected.,Results,2,The protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP were lowered significantly in the high-dose group and modern medicine group compared with those in the model group, and the differences were statistically significant (,P,<,0.05). The ,TNF,-,α, mRNA expression, and the protein and mRNA expression levels of MMP-9, VCAM-1, ICAM-1 and TIMP were lowered in the low-dose and medium-dose groups compared with those in the model group, and the differences were statistically significant (,P,<,0.05). Significantly lowered protein expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP as well as the lowered ,TIMP, mRNA expression were detected in the high-dose group, and compared with the protein and mRNA expression levels in the low dose group the differences were statistically significant (,P,<,0.05). The protein expression levels of VCAM-1 and TIMP were significantly lowered in the medium-dose group compared with those in the low-dose group, and the differences were statistically significant (,P,<,0.05). The expression levels of MMP-9, ICAM-1 and TIMP were significantly lowered in the high-dose group compared with those in the medium-dose group, and the differences were statistically significant (,P,<,0.05).,Conclusion,2,ZGTMG can significantly lower the protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP in the plaque tissues of diabetic AS rats, decrease the expression of inflammatory factors and improve the inflammatory response of vascular endothelium, and the clinical effect of the high-dose group is better than that of the medium-dose and low-dose groups.
