目的,2,观察促愈熏洗方外用对大鼠模拟肛瘘术后创面的愈合作用,并检测创面组织及血清中转化生长因子β1(TGF-β1)、血管内皮生长因子(VEGF)、碱性成纤维细胞因子(bFGF)的表达情况,初步探讨促愈熏洗方促进创面愈合作用的机制。,方法,2,选取54只SD大鼠,制备模拟肛瘘术后创面模型,随机分成模型组、对照组、实验组,每组18只。造模后分别予0.9%NaCl溶液、乳酸依沙吖啶(利凡诺)溶液、促愈熏洗方外用换药处理,每日1次,连续14 d。分别于第3天、第7天、第14天观察各组创面的愈合率,Western blot检测创面组织中TGF-β1、VEGF、bFGF蛋白的表达水平,ELISA法检测血清中TGF-β1、VEGF、bFGF表达的变化。,结果,2,①创面愈合率:3个时点实验组创面愈合率均优于模型组(,P,<,0.05),实验组第3天优于对照组(,P,<,0.01)。②Western blot,结果,2,3个时点实验组创面组织中TGF-β1的表达高于模型组(,P,<,0.01)及对照组(,P,<,0.05)。3个时点各组创面组织中VEGF的表达差异均无统计学意义(,P,>,0.05)。3个时点实验组创面组织中bFGF的表达均略高于对照组与模型组,第3天实验组与对照组均高于模型组(,P,<,0.05,,P,<,0.01),第7天、第14天各组表达的差异无统计学意义(,P,>,0.05)。③ELISA,结果,2,3个时点实验组血清中TGF-β1的表达均高于模型组(,P,<,0.05),实验组第3天高于对照组(,P,<,0.05)。3个时点各组血清中VEGF的表达差异均无统计学意义(,P,>,0.05)。3个时点各组血清中bFGF的表达,第3天表达差异无统计学意义(,P,>,0.05),第7天实验组高于模型组(,P,<,0.01)与对照组(,P,<,0.05),第14天实验组高于模型组(,P,<,0.01)与对照组(,P,>,0.05)。,结论,2,促愈熏洗方能提高大鼠模拟肛瘘术后创面早期的愈合率,其促进创面愈合的机制可能与提高创面组织及血清中TGF-β1、bFGF的表达有关。
Objective,2,To observe the healing rate of wound surface with external application of Cuyuxunxi Prescription after operation of simulated anal fistula in rats, and to detect the expression of transforming growth factor β1 (TGF-β1), vascular endothelial growth factor (VEGF) and basic fibroblast factor (bFGF) in wound tissue and serum, and to explore the mechanism of promoting wound healing.,Methods,2,Fifty-four SD rats were selected to prepare the model of wounds after simulated anal fistula operation and were randomly divided into model group, control group and experimental group, with 18 rats in each group. After the establishment of the model, 0.9% saline, rivanol and Cuyuxunxi Prescription were respectively given for dressing change once a day for 14 days. The wound healing rate of each group was observed on the 3rd, 7th, and 14th days. The expression levels of TGF-β1, VEGF and bFGF in wound tissue were detected by Western blot (WB), and the expression changes of serum TGF-β1, VEGF and bFGF were detected by ELISA.,Results,2,①Wound healing rate: At three time points, the wound healing rate of the experimental group was better than that of the model group (all ,P,<,0.05), and the rate in the experimental group was better than that in the control group on the 3rd day (,P,<,0.01). ②WB results: The expression of TGF-β1 in the wound tissue of the experimental group was higher than that of the model group (,P,<,0.01) and the control group (,P,<,0.05) at 3 time points. There was no statistical significance in the expression of VEGF in the wound tissue of each group at 3 time points (,P,>,0.05). The expression of bFGF in the wound tissue of the experimental group was higher than that of the control group and the model group at 3 time points. The experimental group and the control group were higher than the model group in the expression of bFGF on the 3rd day (,P,<,0.05, ,P,<,0.01), but there was no statistical significance on the 7th,and 14th days (,P,>,0.05). ③ ELISA results: At three time points, the expression of serum TGF-β1 in the experimental group was higher than that in the model group (,P,<,0.05), and that in the experimental group was higher than that in the control group on the 3rd day(,P,<,0.05). There was no statistical significance in the expression of VEGF in the serum of each group at 3 time points (,P,>,0.05). There was no statistical significance in the expression of bFGF in the serum of each group on the 3rd day (,P,>,0.05), but it was higher in the experimental group on 7th day than that in the model group (,P,<,0.01) and the control group (,P,<,0.05) on the 7th day, and higher in the experimental group on the 14th day than that in the model group (,P,<,0.01) and the control group (,P,>,0.05) on the 14th day.,Conclusions,2,Cuyuxunxi Prescription can improve the wound healing rate in the early stage after simulated anal fistula operation in rats. The mechanism of promoting wound healing may be related to the increase of the expression of TGF-β1 and bFGF in wound tissue and serum.
