目的,2,探究熄风制动汤对脂多糖(LPS)诱导的小鼠小胶质细胞(BV-2细胞)炎症反应的作用。,方法,2,将BV-2细胞设置为Control组、LPS组及熄风制动汤组。Control组加入达尔伯克改良伊格尔(DMEM)培养基,LPS组以0.1 mg/L浓度对BV-2细胞进行诱导炎症反应,熄风制动汤组分别以浓度为12.5、25、50、100、200、400 mg·L,-1,的熄风制动汤进行相应干预。采用CCK-8法观察熄风制动汤对BV-2细胞活力的影响;ELISA法检测细胞上清中一氧化氮(NO)水平;real-time PCR检测BV-2细胞炎症因子水平的表达;Western blot法及免疫荧光染色法检测一氧化氮诱导合酶(iNOS)、精氨酸-1(Arg-1)蛋白的表达。,结果,2,熄风制动汤不同剂量组或LPS组培养BV-2细胞后,对其活力无明显影响(,P>,0.05)。与Control组比较,LPS组NO的释放显著增加(,P,<,0.001); M1型小胶质细胞产物肿瘤坏死因子-α(,TNF,-,α,)、白介素-6(,IL,-,6,)、白介素-1β(,IL,-,1β,)mRNA表达增加(,P,<,0.001),M2型小胶质细胞产物转化生长因子-β(,TGF,-,β,)mRNA表达降低(,P<,0.05), M1型标志物iNOS的蛋白表达增加(,P,<,0.01),M2型标志物Arg-1蛋白的表达降低(,P<,0.05)。与LPS组比较,熄风制动汤组(100、200、400 mg·L,-1,)NO的释放(,P,<,0.001)抑制,M1型小胶质细胞产物,TNF,-,α,(,P,<,0.001)、,IL,-,6, mRNA(,P<,0.05)表达降低;熄风制动汤组(400 mg·L,-1,)M2型小胶质细胞产物,IL,-,10,、,TGF,-,β, mRNA表达增加(,P,<,0.05);熄风制动汤组(200 mg·L,-1,)M1型标志物iNOS蛋白表达降低(,P,<,0.01),熄风制动汤组(100 mg·L,-1,)M2型标志物Arg-1蛋白表达增加(,P,<,0.05)。,结论,2,熄风制动汤可以抑制LPS诱导的BV-2细胞向M1损伤型转化,促进向M2修复型转化。
Objective,2,To explore the effect of Xifeng Zhidong Decoction on inflammatory reaction of mouse microglia (BV-2 cells) induced by lipopolysaccharide (LPS).,Methods,2,The microglia of mice were divided into three groups: control group, LPS group and Xifeng Zhidong Decoction group. The DMEM medium was added to the control group. LPS was used to induce inflammation on microglia at a concentration of 0.1 mg·L,-1, in the LPS group, and intervention was performed by Xifeng Zhidong Decoction with a concentration of 12.5, 25, 50, 100, 200 and 400 mg·L,-1 ,respectively in the Xifeng Zhidong Decoction group. The effect of Xifeng Zhidong Decoction on the viability of microglia was observed by cell proliferative toxicity test (CCK-8). The level of nitric oxide (NO) in the supernatant was detected by enzyme linked immunosorbent assay (ELISA), and the expression of inflammatory factors in microglia was detected by fluorescence quantitative polymerase chain reaction (real-time PCR). Western blot and immunofluorescence staining were used to detect the expression of nitric oxide-induced synthase (iNOS) and arginine-1 (Arg-1) proteins.,Results,2,There was no significant effect on the activity of microglia in different doses of Xifeng Zhidong group or LPS group (,P,>,0.05). Compared with the control group, the NO content in microglia supernatant of the LPS group increased significantly(,P,<,0.001); the mRNA expression of ,TNF,-,α,, ,IL,-,6,, ,IL,-,1β, of M1 type microglia increased significantly(,P,<,0.001), but the mRNA expression of ,TGF,-,β, of M2 type microglia decreased (,P,<,0.05) ; the protein expression of M1 marker iNOS increased (,P,<,0.01), but the expression of M2 marker Arg-1 protein decreased (,P,<,0.05). Compared with the LPS group, Xifeng Zhidong Decoction at 100, 200, and 400 mg·L,-1, could significantly inhibit the release of NO(,P,<,0.001), decrease the expression of ,TNF,-,α, IL,-,6, mRNA of M1 type microglia(,P,<,0.001, ,P,<,0.05). Xifeng Zhidong Decoction increased the expression of ,IL,-,10, and ,TGF,-,β ,mRNA of M2 type microglia at 400 mg·L,-1,(,P,<,0.05), reduced the expression of M1 type marker iNOS protein at 200 mg·L,-1, (,P,<,0.01), and increased the expression of M2 type marker Arg-1 protein at 100 mg·L,-1, (,P,<,0.05).,Conclusion,2,Xifeng Zhidong Decoction can inhibit the transformation of microglia to M1 damaged type induced by LPS, and promote the transformation to M2 repair type.
