目的,2,观察补益气血中药四君子汤、四物汤对体外培养人皮肤成纤维细胞EGF、,EGF, mRNA、VEGFA、,VEGF, mRNA表达的影响。,方法,2,实验分为空白对照组、四君子汤组和四物汤组,空白对照组予含完全DMEM培养液进行细胞培养,四君子汤组、四物汤组分别予含10 μg/L、100 μg/L、1 mg/L、10 mg/L、100 mg/L、1 g/L相应中药冻干粉的完全DMEM培养液进行细胞培养。采用CCK-8法测定四君子汤、四物汤对人皮肤成纤维细胞增殖的影响,确定药物浓度和用药时间,Western blot法检测EGF、VEGFA的表达,RT-PCR检测,EGF, mRNA、,VEGF, mRNA的表达。,结果,2,四君子汤、四物汤能促进人皮肤成纤维细胞增殖,各组在浓度为100 mg/L时,促进细胞增殖效果较明显,且药物干预24 h促细胞增殖效果明显。与对照组比较,四君子汤组、四物汤组EGF、,EGF, mRNA、VEGFA、,VEGF, mRNA表达增加(,P,<,0.05,,P,<,0.01)。与四君子汤组比较,四物汤组EGF、,EGF, mRNA表达降低(,P,<,0.05,,P,<,0.01),VEGFA表达差异无统计学意义(,P,>,0.05),,VEGF, mRNA表达量明显增加(,P,<,0.01)。,结论,2,补益气血中药四君子汤、四物汤可通过促进体外培养人皮肤成纤维细胞增殖,上调,EGF, mRNA、,VEGF, mRNA的表达,促进EGF、VEGFA的合成分泌,进而促进血管再生、肉芽组织生长,加速创面愈合;补气中药四君子汤与补血中药四物汤作用存在差异,这可能与中医气与血的不同作用、气血关系以及脏腑理论有关。
Objective,2,To observe the effects of the Chinese medicine Sijunzi Decoction and Siwu Decoction, two prescriptions for invigorating qi and blood, on the expression of EGF,EGF, mRNA, VEGFA and ,VEGF, mRNA in cultured human skin fibroblasts in vitro.,Methods,2,There were three groups divided in this experiment, this is, blank control group, Sijunzi Decoction group and Siwu Decoction group. The cells in the blank control group was cultured with complete DMEM culture medium, and the cells in the Sijunzi Decoction group and Siwu Decoction group were cultured with complete DMEM medium respectively contained 10 μg/L, 100 μg/L, 1 mg/L, 10 mg/L, 100 mg/L, 1 g/L corresponding Chinese medicine freeze-dried powder. CCK-8 method was used to determine the effects of Sijunzi Decoction and Siwu Decoction on the proliferation of human skin fibroblasts, and determine the drug concentration and medication time. Western blot method was used to detect the expression of EGF and VEGFA, and RT-PCR to detect the expression of ,EGF, mRNA and ,VEGF, mRNA.,Results,2,Sijunzi Decoction and Siwu Decoction were able to promote the proliferation of human skin fibroblasts. When the concentration of corresponding Chinese medicine in each group was 100 mg/L, the effect of promoting cell proliferation was obvious, and the effect of drug intervention for 24 h on promoting cell proliferation was quite obvious. Compared with the blank control group, the expression of EGF,EGF, mRNA, VEGFA and ,VEGF, mRNA in Sijunzi Decoction group and Siwu Decoction group was increased (,P,<,0.05,P,<,0.01). Compared with Sijunzi Decoction group, the expression of EGF and ,EGF, mRNA in Siwu Decoction group was decreased (,P,<,0.05,P,<,0.01), and the expression of VEGFA was not significantly different (,P,>,0.05), while the expression of ,VEGF, mRNA was significantly increased (,P,<,0.01).,Conclusions,2,Sijunzi Decoction and Siwu Decoction can promote the proliferation of human skin fibroblasts cultured in vitro, upregulate the expression of ,EGF, mRNA and ,VEGF, mRNA, promote the synthesis and secretion of EGF and VEGFA, and then promote angiogenesis, granulation tissue growth, and accelerate wound healing. There are differences in the effects of Sijunzi Decoction and Siwu Decoction, which may be related to the different effects of qi and blood, the relationship between qi and blood, and the theory of viscera.
