目的,2,观察食道通结方对人食管癌裸鼠移植瘤微血管生成的影响。,方法,2,通过皮下接种人食管癌TE-1细胞株建立人食管癌裸鼠移植瘤模型。将40只荷瘤鼠随机分为模型组、顺铂组、中药组、中药+顺铂组。各组分别予0.9%NaCl溶液、中药食道通结方(18 g·kg,-1,·d,-1,,灌胃,28 d)、顺铂(1 mg·kg,-1,·d,-1,,腹腔注射,d1、d3、d5)、食道通结方(18 g·kg,-1,·d,-1,,灌胃,28 d)+顺铂(1 mg·kg,-1,·d,-1,,腹腔注射,d1、d3、d5)干预。观察各组裸鼠移植瘤的生长情况,免疫组化法检测肿瘤组织CD34的表达,计算肿瘤微血管密度(MVD),免疫组化法检测肿瘤组织血管内皮细胞生长因子A(VEGFA)的表达,采用Western blot法检测肿瘤组织血管内皮细胞生长因子A(VEGFA)、血管内皮细胞生长因子受体2(VEGFR2)、磷酸化血管内皮生长因子受体2(p-VEGFR2)蛋白的表达。,结果,2,与模型组比较,各给药组移植瘤的体积及瘤重均明显减小(,P,<,0.01),以中药+顺铂组的抑瘤作用最强(,P,<,0.05)。与模型组比较,中药组、顺铂组、中药+顺铂组肿瘤MVD及VEGFA、p-VEGFR2的表达均明显降低(,P,<,0.05),且中药+顺铂组下调MVD及VEGFA、p-VEGFR2蛋白表达的作用较顺铂组更为显著(,P,<,0.05)。,结论,2,食道通结方具有拮抗食管癌血管新生的作用,该作用与抑制VEGFA/VEGFR2通路有关。
Objective,2,To observe the effect of Shidao Tongjie Decoction on micro angiogenesis of human esophageal cancer xenografts in nude mice., Methods,2,The xenograft model of human esophageal carcinoma was established by subcutaneous inoculation of human esophageal carcinoma cell line TE-1. Forty tumor bearing mice were randomly divided into 4 groups: model group, Chinese medicine group, cisplatin group and Chinese medicine + cisplatin group. The mice in each group were intervened with 0.9%NaCl, Shidao Tongjie Decoction (18 g·kg,-1,·d,-1, ig, 28 d) , cisplatin (1 mg·kg,-1,·d,-1, ip, d1, d3, d5) , Shidao Tongjie Decoction (18 g·kg,-1,·d,-1, ig, 28 d) + cisplatin (1 mg·kg,-1,·d,-1, ip, d1, d3, d5) , respectively. The growth of transplanted tumor was observed. The expression of CD34 was detected by immunohistochemistry, and microvas cular density (MVD) was calculated according to the expression of CD34. The expressions of vascular endothelial growth factor A (VEGFA) protein was detected by immunohistochemistry.The expressions of VEGFA, vascular endothelial growth factor receptor 2 (VEGFR2) and p-VEGFR2 protein were detected by Western blot.,Results,2,Compared with the model group, the volume and weight of transplanted tumor in treatment groups were significantly reduced (,P,<,0.01) , and the effect of Chinese medicine plus cisplatin was the best (,P,<,0.05) . Compared with the model group, the expressions of MVD, VEGFA and p-VEGFR2 in the tumor tissues of the Chinese medicine group, cisplatin group and the Chinese medicine + cisplatin group decreased significantly (,P,<,0.05) , and the effects of the Chinese medicine + cisplatin group on the expression of MVD, VEGFA and p-VEGFR2 were more significant (,P,<,0.05) .,Conclusion,2,Shidao Tongjie Decoction has the effect of anti-angiogenesis of esophageal cancer, which is related to the inhibition of VEGFA/VEGFR2 pathway.
