目的,2,采用热、药分离的方式探讨明目解毒方熏蒸对于单纯疱疹病毒性角膜炎(HSK)的作用途径。,方法,2,90只健康新西兰大白兔采用随机数字表法分为正常对照组、模型组、中药熏蒸组、热熏蒸组和熏蒸液组5组,每组18只。除正常对照组外,其余各组均采用眼表划痕法给予单纯疱疹病毒I型(HSV-1)接种建立HSK模型。造模后模型组和正常对照组不予药物处理,中药熏蒸组采用明目解毒方每日熏蒸干预,热熏蒸组采用蒸馏水熏蒸,熏蒸液组采用明目解毒方熏蒸液滴眼。分别于造模前1 d和造模1 d、3 d、7 d,对各组实验兔的角膜病变程度进行评分。并在治疗1 d、3 d、7 d随机抽取每组6只眼,采用PCR法检测角膜组织中HSV-1的表达,ELISA法检测房水中IL-2和IFN-γ的含量;采用Western blot方法检测外周血中CD4、CD8的蛋白表达情况。,结果,2,治疗3 d、7 d,中药熏蒸组、熏蒸液组和热熏蒸组的兔角膜HSV-1含量低于模型组(,P,<,0.05);各组间房水中IFN-γ的含量差异无统计学意义(,P,>,0.05),治疗7 d,中药熏蒸组和热熏蒸组存在交互作用。治疗1 d、3 d、7 d,中药熏蒸组房水中IL-2含量与模型组差异有统计学意义(,P,<,0.05);治疗1 d、7 d熏蒸液组与模型组房水中IL-2含量差异有统计学意义(,P,<,0.05)。治疗3 d、7 d中药熏蒸组较模型组外周血CD4蛋白表达明显增强(,P,<,0.05)。,结论,2,明目解毒方熏蒸可通过热、药效应清除HSK角膜组织中的HSV-1病毒,并且影响局部免疫因子IL-2水平,上调外周血CD4的表达。
Objective,2,To explore the effect of Mingmu Jiedu Prescription (MJP) fumigation on herpes simplex keratitis(HSK)by means of heat and drug separation.,Methods,2,Ninety healthy New Zealand white rabbits were randomly divided into five groups: model group (,n,=18) , traditional Chinese medicine thermal fumigation group (,n,=18) , thermal fumigation group (,n,=18) , fumigant group (,n,=18) and normal group (,n,=18) .Except for the normal group, all the other groups were inoculated with HSV-1 virus to establish HSK mode by ocular surface scratching method. After modeling, the model group and normal group were not treated with drugs, while the traditional Chinese medicine thermal fumigation group was fumigated with MJP every day, the thermal fumigation group was fumigated with distilled water, and the fumigant group was treated with MJP fumigation liquid eye drops.The degree of keratopathy in each group was evaluated at 1 day before modeling and 1 day, 3 days and 7 days after modeling.On the 1st, 3rd and 7th days after treatment, 6 eyes in each group were randomly selected for detecting the expression of HSV-1 in corneal tissue by PCR method, the contents of IL-2 and IFN-γ in aqueous humor by ELISA method, and the protein expressions of CD4, CD8 in peripheral blood by Western blot method.,Results,2,On the 3rd and 7th days after treatment, the corneal HSV-1 contents in the traditional Chinese medicine thermal fumigation group, the fumigant group and the thermal fumigation group were lower than those in the model group (,P,<,0.05) .But there was no significant difference in the content of IFN-γ in aqueous humor among the three groups (,P,>,0.05) .On the 7th day after treatment, there was interaction between traditional Chinese medicine thermal fumigation group and thermal fumigation group. On the 1st, 3rd and 7th days after treatment, the content of IL-2 in aqueous humor of the traditional Chinese medicine thermal fumigation group was significantly different from that of the model group, and there was significant difference between the fumigant group and the model group on the 1st and 7th days after treatment (,P,<,0.05) .The expression of CD4 protein in peripheral blood in the traditional Chinese medicine thermal fumigation group was significantly higher than that in the model group after 3 and 7 days of treatment (,P,<,0.05) .,Conclusion,2,MJP fumigation treatment can eliminate HSV-1 virus in HSK corneal tissue through thermal and drug effects, affect the level of local immune factor IL-2, and up-regulate the expression of CD4 in peripheral blood.
