Objective:To research the effect of Guanxinkang on the cholesterol level of HepG2 cells induced by ox-LDL at different concentrations,as well as the cholesterol metabolism-related miR-33a,miR-122 and LDLR. MethodsSerum containing Guanxinkang and simvastatin were prepared respectively,and HepG2 cells were randomly divided into blank control group,model group,simvastatin group and Guanxinkang group. HepG2 cells were induced by ox-LDL with concentrations of 20,40,80 and 160 mg/L respectively for 24 hours,and then incubated with serum containing drugs for 48 hours respectively. The cell morphology of HepG2 cholesterol metabolism model was observed by oil red O staining. The expression levels of mir-33a,mir-122 and LDLR genes were detected by real-time PCR,and the protein expression of LDLR was detected by Western blot. Results:Compared with the model group, Guanxinkang can significantly reduce the content of orange lipid droplets in HepG2 cells after ox-LDL induction at different concentrations,up-regulate the expression of LDLR gene and protein in the cells after ox-LDL induction,and inhibit the expression of miR-33a and miR-122 gene(P<0.05, P<0.01). Conclusion:Guanxinkang can up-regulate the expression of LDLR gene and protein,inhibit the expression of miR-33a and miR-122 gene,thereby reducing the content of cholesterol in cells and alleviating the process of cholesterol metabolism imbalance.
