目的:对车前种子和平车前种子的成分差异进行分析,比较其对链脲佐霉素(STZ)诱导的C57BL/6糖尿病模型小鼠的作用,为车前子的品质评价提供实验依据。 方法:采用薄层色谱法,对车前种子和平车前种子的成分进行定性鉴别比较;采用HPLC法,对主要指标活性成分京尼平苷酸和麦角甾苷进行含量测定和比较。同时通过小鼠腹腔注射STZ制备糖尿病模型,药物干预后,测定小鼠空腹血糖,收集血液、胰腺组织,测定血清生化指标,比较两种不同基原车前子对模型小鼠的作用。 结果:化学成分比较分析结果显示,平车前种子与车前种子除共同含有苯乙醇苷类麦角甾苷、环烯醚萜类京尼平苷酸、胍类车前素A外,还含有其他专属性成分;指标成分HPLC含量测定结果表明,车前种子中京尼平苷酸、麦角甾苷的含量明显高于平车前种子。在对实验性糖尿病小鼠的作用观察中,车前种子组和平车前种子组小鼠的体质量显著增加,血糖明显降低,血清总抗氧化能力增强,血清中过表达的TNF-α和IL-1β含量降低(P<0.01);其中车前种子降低小鼠血糖的效果要显著优于平车前种子,并且能提升血清中胰岛素的含量和修复胰岛β细胞的损伤(P<0.05),而平车前种子没有明显作用。 结论:车前种子和平车前种子的化学成分存在差异,车前种子京尼平苷酸、麦角甾苷的含量明显高于平车前种子;车前种子和平车前种子对STZ诱导的糖尿病小鼠模型,均具有良好的降血糖效果,但车前种子作用更佳。
Objective:To analyze the differences in the composition of Plantagoasiatica L.seeds and Plantagodepressa Willd. seeds and compare their therapeutic effects on C57BL / 6 diabetic mice induced by STZ(streptozotocin)in order to provide an experimental basis for the quality evaluation on Plantaginis Semen. MethodsThe composition of Plantagoasiatica L.seeds and Plantagodepressa Willd. seeds were compared qualitatively by TLC. A quantitative method for the determination of geniposidic acid and acteoside was established by HPLC. At the same time,the diabetic model was made by intraperitoneal injection of STZ in mice. After drug intervention,fasting blood glucose was measured,blood and pancreatic tissue were collected,serum biochemical indexes were measured,and the effects of two different primordial plantain on the model mice were compared. Results:The results of chemical composition analysis showed that the seeds of Plantagoasiatica L.and Plantagodepressa Willd. contain not only phenylethanolic glycoside acteoside,iridoid terpenoid geniposidic acid,plantagoamidinic acid A,but also other specific components. The content of geniposidic acid and acteoside in Plantagoasiatica L. seeds was significantly higher than in Plantagodepressa Willd. seeds by HPLC. In the experimental diabetic mice,the weight of the mice was significantly increased in both groups of Plantagoasiatica L.seeds and Plantagodepressa Willd. seeds,the blood glucose was significantly reduced,the total serum antioxidant capacity was enhanced,and the overexpressed TNF-α and IL-1β in serum was decreases(P<0.01). Among them, Plantagoasiatica L.seeds could significantly reduce blood glucose of mice than Plantagodepressa Willd. seeds, and could increase the content of insulin in serum and repair the damage of islet β cells(P<0.05),while Plantagodepressa Willd. seeds had no obvious effect. Conclusion:The chemical constituents of Plantagoasiatica L. seeds and Plantagodepressa Willd. seeds are different and the contents of geniposidic acid and acteoside in Plantagoasiatica L.seeds are higher than those of Plantagodepressa Willd. seeds. Both Plantagoasiatica L. seeds and Plantagodepressa Willd. seeds have good effects on STZ induced diabetic mice model. However,the effect of Plantagoasiatica L. seeds is better than that of Plantagodepressa Willd. seeds.
