摘要

目的：探讨益气复生方对人胃癌细胞BGC-823增殖和迁移的影响。方法：采用MTT法检测益气复生方对人胃癌细胞BGC-823活性的抑制作用；划痕实验检测益气复生方对细胞迁移的影响；流式细胞术检测益气复生方对细胞周期和凋亡的影响。结果：与空白对照组比较,不同浓度(0.05 mg/mL、0.1 mg/mL、0.2 mg/mL、0.4 mg/mL、0.8 mg/mL、1 mg/mL)益气复生方作用的细胞,细胞活性降低；划痕结果显示益气复生方处理的细胞迁移能力明显低于空白对照组；细胞周期检测显示益气复生方能阻滞人胃癌细胞BGC-823的周期进展；细胞凋亡检测表明益气复生方能诱导人胃癌细胞BGC-823凋亡。结论：益气复生方能有效抑制人胃癌细胞BGC-823的增殖和迁移,从而起到抗胃癌BGC-823细胞生长的作用。

Abstract

Objective:To investigate the effect of Yiqifusheng prescription on the proliferation and migration of human gastric cancer cell line BGC-823. MethodsMTT assay was used to detect the inhibitory effect of Yiqifusheng prescription on BGC-823 cell viability. The effect of Yiqifusheng prescription on cell migration was tested by wound healing test in BGC-823 cell. Flow cytometry was used to determine the effects of Yiqifusheng prescription on cell cycle and apoptosis. Results:Compared to the blank control group, BGC-823 cells treated with various concentrations of Yiqifusheng prescription (0.05 mg/mL,0.1 mg/mL,0.2 mg/mL,0.4 mg/mL,0.8 mg/mL,1 mg/mL) decreased cell activity. The wound healing test results showed that the BGC-823 cell migration ability of Yiqifusheng prescription-treated group was reduced than that of the blank control group. Cell cycle test showed that Yiqifusheng prescription could block the cycle progression of human gastric cancer cells BGC-823.Apoptosis test showed that Yiqifusheng prescription could promote cell apoptosis of human gastric cancer cells. Conclusion:Yiqifusheng prescription can significantly inhibit the proliferation and migration of human gastric cancer cell,and play an anti-growth role in gastric cancer cells of BGC-823.
