目的:研究黄连-大黄-肉桂复方及其最佳组分配伍对肝癌小鼠肿瘤增殖及证候的影响。 方法:通过右腋下皮下注射人肝癌细胞建立移植瘤裸鼠肝癌模型,成模小鼠分为模型对照组(模型组)、黄连-大黄-肉桂复方组(复方组)、小檗碱-大黄素-桂皮醛组方组(组方组),各组给予相应的药物治疗,共2周。给药期间观察小鼠的状态,给药前后分别测定各组小鼠的瘤径并计算瘤体积,测定体质量、腋温、抓力、旷场水平移动格数及垂直站立次数、爪尾显微拍照,据此计算各组的阴盛衰度、血盛衰度、气盛衰度和阳盛衰度,以各四诊指标治疗后/治疗前的比值用于分析证候的变化。最后取瘤体称重,计算抑瘤率。 结果:①小鼠状态:实验过程中,各组小鼠状态尚可,活动较灵敏,大小便正常。②抑瘤作用:各组小鼠瘤体积随时间增大,其中模型组>复方组>组方组,瘤重结果与瘤体积一致。③证候变化:两用药组小鼠的气盛衰度均有所提升,组方组优于复方组;两用药组小鼠的血盛衰度均有所升高,复方组优于组方组;两用药组小鼠的阴盛衰度、阳盛衰度均变化不大。 结论:黄连-大黄-肉桂复方及其最佳组分配伍均能一定程度抑制肝癌小鼠肿瘤的增殖,且具有益气血的趋势。
Objective:To explore the effects of Huanglian(Rhizoma Coptidis)-Dahuang(Radix et Rhizoma Rhei)-Rougui(Cortex Cinnamomi)compound and its best components compatibility on tumor proliferation and syndromes of liver cancer mice. MethodsHuman hepatocarcinoma cells were injected subcutaneously into the right axilla to establish a liver cancer model of transplanted nude mice.After modeling success,mice were divided into model control group(model group),Huanglian(Rhizoma Coptidis)-Dahuang(Radix et Rhizoma Rhei)-Rougui(Cortex Cinnamomi)compound group(compound group) and berberine-emodin-cinnamaldehyde group (compatibility group). Each group was treated with corresponding drug for 2 weeks.During the administration, the state of the mice was observed.Before and after administration,the tumor diameter and volume,body weight, axillary temperature, grip force, number of horizontal movements and vertical standings in the squat field, microphotograph of the claws and tails were measured twice.The degree of excess or deficiency of Qi,Xue,Yin and Yang were calculated according to the above indexes.The ratios of post-treatment/pre-treatment of these indexes were used to analyze the change of syndromes.Finally, the mice were sacrificed and the tumors were weighed and the tumor inhibition rate was measured. Results:①Mice state:During the experiment, the mice were all in good condition, both the movement and the defecation were normal.②Antitumor effect:The tumor volume of each group increased with time,which was displayed as the model group> compound group>compatibility group. The variation of tumor weight was consistent with that of the tumor volume.③Syndrome change:After treatment,the degree of excess or deficiency of Qi was increased,and the degree of the compatibility group was better than that of the compound group.The degree of excess or deficiency of Xue was also raised,and the degree of the compound group was superior to the compatibility group.The degree of excess or deficiency of Yin and Yang almost had no change. Conclusion:Both the compound and the best compatibility of the medicines can inhibit the tumor proliferation of liver cancer mice and has the trend of invigorating qi and blood.
