目的:探讨六味地黄丸对甲状腺素诱导的阴虚证小鼠肝脏损伤的保护作用。 方法:ICR系小鼠30只随机分为正常组、模型组和六味地黄丸组(生药含量1.8 g /kg),除正常组外其他小鼠用皮下注射L-甲状腺素(280 μg/kg)方法建立阴虚证模型。1周后,造模同时开始灌胃给予六味地黄丸溶液,连续3周,其间测量小鼠体质量、体温、自主活动次数、痛阈和心率,评价阴虚证模型以及药物的对抗作用。实验结束时检测血清中丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)活性,以及总蛋白(TP)、总胆固醇(TC)和三酰甘油(TG)含量;剥离肝脏并称重,计算肝脏指数;HPLC法检测肝脏能量物质磷酸肌酸(PCr)、三磷酸腺苷(ATP)、二磷酸腺苷(ADP)和单磷酸腺苷(AMP)含量,计算腺苷酸总含量TAN(ATP+ADP+AMP);检测肝脏环磷腺苷(cAMP)含量。 结果:模型小鼠体质量、体温、自主活动次数、痛阈和心率等阴虚指标出现异常,六味地黄丸有一定的改善作用。与正常组比较,模型组肝脏指数和血清ALT和AST活性显著升高,血清TC和TP水平显著降低,肝脏组织 ATP、ADP、AMP、cAMP、TAN含量显著降低(P<0.05,P<0.01)。与模型组比较,六味地黄丸组血清ALT和AST活性显著降低,血清TC和TP水平显著升高,肝脏组织ADP、cAMP、TAN含量显著升高(P<0.05,P<0.01),ATP、AMP含量也有升高趋势。 结论:六味地黄丸对L-甲状腺素诱导的阴虚证小鼠肝脏损伤具有一定的保护作用,这可能与改善肝脏组织能量代谢紊乱作用有关。
Objective:To investigate the protective effect of Liuwei Dihuang pill on liver injury in mice with yin deficiency induced by thyroxine. MethodsThirty ICR mice were randomly divided into normal group, model group and Liuwei Dihuang pill group (1.8 g crude drug/kg). Except for the normal group, mice in the other groups were injected subcutaneously with L-thyroxine (280 μg/kg) solution to establish the model of yin deficiency. One week later, the solution of Liuwei Dihuang pill was given by intragastric administration, while L-thyroxine was continued to be administered subcutaneously for 3 weeks. During this period, body mass, temperature, number of locomotor activities, pain threshold and heart rate of the mice were measured to evaluate the yin deficiency model and antagonistic effect of the drug. At the end of experiment, the serum activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and contents of total protein (TP), total cholesterol (TC) and triglyceride (TG) were determined; the liver was dissected and weighed to calculate the liver index; the contents of phosphocreatin (PCr), adenosine triphosphate (ATP), adenosine monophosphate (ADP), adenosine monophosphate (AMP) and cyclic adenosine monophosphate (cAMP) in liver were determined by HPLC and the total adenylate content TAN (ATP+ADP+AMP) was calculated. Results:The yin deficiency indicators such as body mass, temperature, number of locomotor activities, pain threshold and heart rate of the model mice were abnormal, and Liuwei Dihuang pill showed certain effect in improving these Compared with the normal group, the liver index and the serum activities of ALT and AST increased significantly, the serum levels of TC and TP decreased significantly and the contents of ATP, ADP, AMP, cAMP and TAN in liver tissues decreased significantly in the model group(P<0.05,P<0.01). Compared with the model group, the serum activities of ALT and AST decreased significantly in the Liuwei Dihuang pill group, the serum levels of TC and TP increased significantly, and in liver tissues, the contents of ADP, cAMP and TAN increased significantly(P<0.05,P<0.01), and the contents of ATP and AMP also increased in the Liuwei Dihuang pill group. Conclusion:Liuwei Dihuang pill has a certain protective effect on liver injury induced by L-thyroxine in mice with yin deficiency, which may be related to the improvement of energy metabolism disorder in liver tissues.
