目的:探究清营汤加减对急性湿疹大鼠背部皮肤TNF-α、IL-4表达的影响及作用机制。 方法:SD大鼠60只随机分为6组:氯雷他定组,清营汤高、中、低剂量组,模型组和正常组,每组10只。除正常组外,剩余各组对大鼠皮肤右侧背部,使用2,4-二硝基氯苯(DNCB)试剂造模,给予清营汤加减药液、氯雷他定药液、0.9%NaCl溶液对造模成功大鼠进行灌胃治疗。治疗后对实验组大鼠湿疹严重程度进行评分评估;运用称重法比较治疗后皮肤的重量差异;并采用免疫组化法(SP法)检测大鼠右侧背部皮肤组织中TNF-α、IL-4含量。 结果:①与模型组比较,清营汤灌胃中、低剂量组实验大鼠湿疹程度评分均明显降低(P<0.05),高剂量组降低尤为显著(P<0.01),说明清营汤加减可明显改善DNCB所致实验大鼠皮肤糜烂、水肿、红斑症状。②与模型组比较,氯雷他定组、清营汤灌胃组实验大鼠皮肤肿胀程度显著减轻(P<0.01)。与清营汤低剂量组比较,清营汤中、高剂量组肿胀度明显减轻(P<0.05)。与清营汤中剂量组比较,清营汤高剂量组肿胀度明显减轻(P<0.05)。与氯雷他定组比较,清营汤灌胃高剂量组大鼠皮肤肿胀程度明显减轻(P<0.05)。③与模型组比较,氯雷他定组、清营汤灌胃组大鼠病变皮肤组织中TNF-α、IL-4含量明显下降(P<0.01);与氯雷他定组比较,清营汤灌胃高剂量组实验大鼠病变皮肤组织中TNF-α、IL-4含量显著降低(P<0.05)。 结论:清营汤加减治疗急性湿疹可明显改善渗出、糜烂、红斑、肿胀等症状,其治疗机制可能为通过调控TNF-α、IL-4的水平来发挥治疗作用。
Objective:To investigate the effects of modified Qingying Decoction on the expression of TNF-α and IL-4 in dorsal skin of rats with acute eczema. MethodsSixty SD rats were randomly divided into 6 groups, 10 rats in each group:loratadine group, high-, middle-, and low-dose Qingying Decoction groups, model group and normal group. No modelling treatment was performed in the normal group. Rats in the other groups were modeled by applying 2,4-Dinitrochlorobenzene (DNCB) reagent on the right back of rats’ skin, and then modified Qingying Decoction, loratadine solution, and 0.9% NaCl solution were given to the successfully modelled rats by gastric gavage. After treatment, the severity of eczema in the experimental groups was evaluated by scoring. Skin weight was measured and compared after treatment. The content of TNF-α and IL-4 in skin tissue of the right back of rats was determined by immunohistochemistry (SP method). Results:①Compared with the model group, the scores of severity of eczema were reduced significantly in the middle- and low-dose Qingying Decoction groups (P<0.05), especially in the high-dose group (P<0.01), suggesting that Qingying Decoction could significantly improve the symptoms of skin erosion, edema and erythema in rats caused by DNCB. ②Compared with the model group, the degree of swelling was reduced significantly in the loratadine group and the Qingying Decoction groups (P<0.01); it was reduced significantly in the middle- and high-dose Qingying Decoction groups compared with the low-dose group (P<0.05); it was reduced significantly in the high-dose Qingying Decoction group compared with the middle-dose group (P<0.05); it was reduced significantly in the high-dose Qingying Decoction group compared with the loratadine group (P<0.05). ③Compared with the model group, the content of TNF-α and IL-4 in the affected skin tissue decreased significantly in the loratadine group and the Qingying Decoction groups (P<0.01); it decreased significantly in the high-dose Qingying Decoction group compared with the loratadine group (P<0.05). Conclusion:Modified Qingying Decoction can significantly improve the symptoms of local exudation, erosion, erythema and swelling in rats with acute eczema and it may exert the therapeutic effects by regulating the levels of TNF-α and IL-4.
