目的:研究肝郁气滞血瘀证模型大鼠下丘脑及胃内饥饿素(ghrelin)表达的变化以及护肝方919糖浆的干预作用。 方法:40只Wistar大鼠随机分为正常组,模型组,919糖浆高、中、低剂量组(27、13.5、6.75 g·kg-1),每组8只。除正常组外,其余各组采用夹尾急性激怒法制作肝郁气滞血瘀证模型,各组给予相应的干预措施,造模21天。其间监测各组大鼠的摄食量及体质量。实验结束后采用ELISA法测定各组大鼠血清酰基化ghrelin浓度,Western blot法测大鼠胃及下丘脑ghrelin蛋白表达,RT-PCR法检测胃中ghrelin、生长激素促分泌受体(GSHR)、5-羟色胺2b受体(5-HT,2b,R)mRNA表达以及下丘脑ghrelin、5-羟色胺2c受体(5-HT,2c,R)mRNA水平。 结果:与正常组比较,模型组大鼠血清酰基化ghrelin水平显著升高(P,<,0.01),下丘脑及胃ghrelin蛋白表达增加(P,<,0.01),胃内ghrelin、GHSR及下丘脑ghrelin mRNA表达水平显著上调(P,<,0.01),胃内5-HT,2b,R及下丘脑内5-HT,2c,R mRNA水平显著下调(P,<,0.01)。与模型组比较,919糖浆治疗组能显著逆转模型大鼠血清、胃内及下丘脑ghrelin水平的升高(P,<,0.01),下调模型大鼠胃内GHSR mRNA水平(P,<,0.01),上调胃内5-HT,2b,R及下丘脑内5-HT,2c,R mRNA水平(P,<,0.01)。 结论:ghrelin是肝郁气滞血瘀证的重要物质基础;919糖浆能通过调节模型大鼠脑胃ghrelin通路的异常表达,从而发挥其防治肝郁气滞血瘀证的作用。
Objective:To investigate the changes in the expression of ghrelin in hypothalamus and stomach and the effects of liver-protective 919 syrup on it in rats with liver-depression, qi-stagnation and blood-stasis syndrome. Methods40 Wistar rats were randomly assigned into normal group, model group, and 919 syrup high-, medium-and low-dose groups (27,3.5 and 6.75 g·kg-1 respectively), 8 rats in each group. Excluding the normal group, the rat model of liver-depression, qi-stagnation and blood-stasis was established by acute tail entrapment method. The groups were given corresponding intervention for modelling for 21 days. Food intake and body mass of rats in each group were monitored during the process. After experiment, the serum acylated ghrelin concentrations in each group were determined by ELISA, and the ghrelin protein expression in stomach and hypothalamus was detected by Western blot. The mRNA expression of ghrelin, growth hormone secretagogue receptor (GSHR) and 5-hydroxytryptamine 2b receptor (5-HT,2b,R) in the stomach and the mRNA expression of ghrelin and 5-HT,2c,R in the hypothalamus were measured by RT-PCR. Results:Compared to the normal group, the level of serum acylated ghrelin was significantly increased (P,<,0.01), the expression of ghrelin protein in hypothalamus and stomach was increased (P,<,0.01), the mRNA expression ghrelin and GHSR in stomach and of ghrelin in hypothalamus were significantly increased (P,<,0.01), and the mRNA level of 5-HT,2b,R in stomach and 5-HT,2c,R in hypothalamus was significantly reduced (P,<,0.01) in the model group. Compared to the model group, 919 syrup could significantly reverse the increases in the level of ghrelin in serum, stomach and hypothalamus in model rats (P,<,0.01), downregulate the mRNA level of GHSR in stomach and upregulate the mRNA levels of 5-HT,2b,R in stomach and 5-HT,2c,R in hypothalamus in model rats (P,<,0.01). Conclusion:Ghrelin is an important material basis of liver-depression, qi-stagnation and blood-stasis; 919 syrup is effective in preventing and treating the syndrome by regulating the abnormal expression of ghrelin in hypothalamus and stomach.
