目的:研究羟喜树碱对人前列腺癌PC-3细胞增殖抑制作用及能量代谢和关键酶的影响。 方法:选取PC-3细胞,采用MTT法测定1.0、2.0及5.0 μmol/L的羟喜树碱处理该细胞24 h后细胞的增殖抑制能力。采用分光光度法测定葡萄糖摄取量及乳酸生成量及ATP生成相关能量代谢参数。采用Western blot法测定乳酸脱氢酶A(LDHA)和AMP活化蛋白激酶(AMPK)表达。 结果:1.0、2.0及5.0 μmol/L的羟喜树碱可抑制PC-3细胞的增殖并呈剂量依赖性(P,<,0.05,P,<,0.01),IC,50,值为3.24 μmol/L。与对照组相比,不同浓度的羟喜树碱可降低葡萄糖摄取,抑制乳酸生成及ATP生成,且均呈剂量依赖性(P,<,0.05,P,<,0.01);Western blot结果显示,不同浓度的羟喜树碱可使PC-3细胞中LDHA及AMPK表达降低(P,<,0.05,P,<,0.01)。 结论:羟喜树碱通过可降低PC-3细胞中LDHA及AMPK表达,抑制肿瘤的有氧酵解并抑制细胞增殖。
Objective:To investigate the effect of hydroxycamptothecin on proliferation and energy metabolism of human prostate cancer PC-3 cells and on the critical enzymes in these cells. MethodsUsing MTT method, the proliferation inhibitory ability was determined after the PC-3 cells were treated with hydroxycamptothecin at 1.0,2.0 and 5.0 μmol/L for 24h. The energy metabolism parameters related to glucose uptake, lactic acid production and ATP production were determined with spectrophotometry. The expression of lactate dehydrogenase A (LDHA) and AMP-activated protein kinase (AMPK) was detected by Western blotting. Results:Hydroxycamptothecin inhibited proliferation of the PC-3 cells at 1.0,2.0 and 5.0 μmol/L in a dose-dependent manner (P<0.05, P<0.01), with an IC50 value of 3.24 μmol/L. Compared with the control group, hydroxycamptothecin at different concentrations could reduce glucose uptake, inhibit lactic acid production and ATP production in a dose-dependent manner (P<0.05, P<0.01). Western blotting results showed that hydroxycamptothecin at different concentrations could reduce the expression of LDHA and AMPK in PC-3 cells (P<0.05, P<0.01). Conclusion:Hydroxycamptothecin inhibits aerobic glycolysis and cell proliferation in the tumor by reducing the expression of LDHA and AMPK in PC-3 cells.
