目的:观察鹿芪颗粒对急性心肌梗死大鼠心功能及血清内毒素(LPS)水平的影响。 方法:60只雄性成年健康Wistar系大鼠随机分为假手术组、模型组、万古霉素组(0.075 g·300 ml-1·d-1)、中药低剂量组(0.25 ml·kg-1·d-1)、中药中剂量组(0.5 ml·kg-1·d-1)、中药高剂量组(0.75 ml·kg-1·d-1),每组10只。除假手术组外,其余5组大鼠通过结扎冠状动脉左前降支建立心肌梗死模型,假手术组大鼠除不结扎冠状动脉外,余手术方法相同。造模成功后各药物组分别给予相应药物,假手术组、模型组给予等量0.9%NaCl溶液,连续干预6周。观察各组大鼠心功能、血清LPS水平及心肌组织病理学情况。 结果:①=1\*GB3与假手术组比较,模型组LVEF、FAS值降低(P<0.05),血清LPS水平升高(P<0.05)。②=2\*GB3与模型组比较,中药中、高剂量组大鼠LVEF、FAS值升高(P<0.05),③=3\*GB3与万古霉素组比较,中药中、高剂量组大鼠LVEF及FS值升高(P<0.05),④=4\*GB3与中药低剂量组比较,中药中、高剂量组大鼠LVEF及FS值升高(P<0.05),血清LPS水平降低(P<0.05)。⑤=5\*GB3中药中、高剂量组组间比较,大鼠心功能、血清LPS水平差异无统计学意义(P>0.05)。 结论:鹿芪颗粒可改善急性心肌梗死大鼠的心功能,减少心肌组织炎性面积,其机制可能与降低血清LPS水平、维持心脏稳态有关。
Objective:To observe the effect of Luqi Granule on cardiac function and serum endotoxin (lipopolysaccharid, LPS) level in rats with acute myocardial infarction (MI). Methods60 healthy adult male Wistar rats were randomly assigned into sham operation group, model group and vancomycin group (0.075 g·300 ml-1·d-1), low-dose Chinese medicine group (0.25 ml·kg-1·d-1), middle-dose Chinese medicine group (0.5 ml·kg-1·d-1) and high-dose Chinese medicine group (0.75 ml·kg-1·d-1), 10 rats in each group. Except the sham operation group, MI model was established with rats in the other 5 groups by ligating the left anterior descending coronary artery. The sham operation group received the same procedure except that the coronary artery was not ligated. After successful modeling, each drug group was given the corresponding drug, and the sham operation group and the model group were given an equal volume of 0.9% NaCl solution for 6 weeks. Cardiac function [left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (FS)], serum LPS level and myocardial histopathology were observed. Results:①=1\*GB3Compared with the sham operation group, the model group had lower LVEF and FS values (P<0.05) and a higher serum LPS level (P<0.05). ②=2\*GB3Compared with the model group, the serum LPS level decreased in the vancomycin group and the middle- and high-dose Chinese medicine groups (P<0.05); the LVEF and FS values increased in the middle- and high-dose Chinese medicine groups (P<0.05). ③=3\*GB3Compared with the vancomycin group, the LVEF and FS values increased in the middle- and high-dose Chinese medicine groups (P<0.05). ④=4\*GB3Compared with the low-dose Chinese medicine group, the LVEF and FS values increased and the serum LPS level decreased in the middle- and high-dose Chinese medicine groups (P<0.05). ⑤=5\*GB3There were no significant differences in cardiac function and serum LPS level between the middle- and high-dose Chinese medicine groups (P>0.05). Conclusion:Luqi granule improves the cardiac function of rats with acute MI and reduces inflammatory area of myocardial tissue. The mechanism may be related to decrease in serum LPS level and maintenance of cardiac homeostasis.
