Objective:To study the effects on body mass and blood leptin in offspring after intervention of mice under postpartum immobilization stress with Huganfang 919 syrup. MethodsThe postpartum female mice were randomly assigned into the control group, the immobilization stress (IS) group and the 919 syrup treatment group. At the time of IS, the 919 syrup treatment group was administered 919 syrup by gastric gavage daily (27 g·kg-1·d-1), the IS group was given an equal dose of 0.9% NaCl solution and the control group received no treatment. At weaning 3 weeks later, changes in the body mass, blood leptin and Ob-Rb mRNA in the liver were observed in the offspring. Results:Compared to the control group, decreased body mass was observed in the IS and the 919 syrup treatment groups at 6,3, 18 and 23 weeks of age, with statistically significant difference (P<0.05); and there was no statistically significant difference in body mass between the IS and the 919 syrup treatment groups (P>0.05). At 23 weeks of age, the level of plasma leptin increased in the IS group compared to the control group, while it decreased in the 919 syrup treatment group compared to the IS group, both with statistically significant difference (P<0.01). At 23 weeks of age, the relative expression of Ob-Rb mRNA in the liver increased significantly in the IS group compared to the control group, (P<0.01), while it decreased significantly in the 919 syrup treatment group compared to the IS group (P<0.05); and there was no significant difference in the male offspring among the three groups (P>0.05). Conclusion:The low body mass caused by postpartum IS is associated with leptin, and Huganfang 919 syrup improves the changes in leptin by regulating OB-Rb mRNA in the liver.
