目的:观察补阳还五汤、少腹逐瘀汤和丹参饮3首活血化瘀方药对高脂血症模型大鼠红细胞膜生物学特征的影响,明确三方均高脂血症在红细胞膜上的作用靶点及作用机制。 方法:SD大鼠90只,雌雄各半,随机分为补阳还五汤高剂量组(生药含量14.0 g·kg-1)、低剂量组(生药含量3.6 g·kg-1),丹参注射液高剂量组(生药含量3.6 g·kg-1)、低剂量组(生药含量0.9 g·kg-1),少腹逐瘀汤高剂量组(生药含量4.8 g·kg-1)、低剂量组(生药含量1.2 g·kg-1),阳性对照药辛伐他汀组(2.0 mg·kg-1),以及模型组和空白组。以灌胃给予脂乳剂复制高脂血症大鼠模型,连续20天,之后分别灌胃给予相应药物治疗10天,治疗同时继续灌胃脂乳剂维持高脂血症状态。取血测定血液流变学及红细胞膜组分相关指标。 结果:与模型组比较,三方对血浆中TG、TC、LDL-C、GLU的含量均可显著降低(P,<,0.05,P,<,0.01),对HDL-C的含量显著升高(P,<,0.05,P,<,0.01);三方都可降低全血黏度和血浆黏度(P,<,0.05,P,<,0.01);三方均可显著延长APTT时间,降低FIB含量(P,<,0.05,P,<,0.01),丹参饮和补阳还五汤可显著延长PT(P,<,0.05)。三方均能显著升高巯基含量及SOD活性(P,<,0.05,P,<,0.01);此外,丹参饮显著升高Na,+,-K,+,-ATP酶活性,降低MDA含量(P,<,0.05,P,<,0.01),补阳还五汤显著升高Na,+,-K,+,-ATP酶活性和唾液酸含量(P,<,0.05,P,<,0.01);少腹逐瘀汤显著升高唾液酸含量(P,<,0.01)。 结论:三方均具有活血化瘀降血脂的作用。三方共同点是可能通过提高清除自由基和抗氧化能力来达到活血化瘀降血脂的作用,此外各方还通过改善红细胞变形性、聚集性等方面改善血瘀状态,从而达到活血化瘀降血脂的作用。
Objective:To observe the effects of Buyang Huanwu Decoction, Shaofu Zhuyu Decoction and Danshen Drink on the biological characteristics of erythrocyte membrane in hyperlipidemia rats, and clarify the target and mechanism of three prescriptions on the erythrocyte membrane in hyperlipidemia. MethodsNinety SD rats, male and female in half, were randomly divided into the Buyang Huanwu Decoction groups with high (14.0 g crude drug·kg-1) and low-dose (3.6 g crude drug·kg-1), Danshen Injection groups with high- (3.6 g crude drug·kg-1) and low-dose (0.9 g crude drug·kg-1), Shaofu Zhuyu Decoction groups with high-(4.8 g crude drug·kg-1) and low-dose (1.2 g crude drug·kg-1), positive control simvastatin group (2.0 mg·kg-1), model group and blank group. The rat model of hyperlipidemia was replicated by intragastric administration of fat emulsion for 20 days. Then each group was treated with the corresponding drug treatment for 10 days, meanwhile, the fat emulsion treatment was continued to maintain the hyperlipidemia state. The blood was collected and the related indexes of hemorheology and the components of erythrocyte membrane were determined. Results:Compared with the model group, the plasma contents of triglyceride (TG), totel cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and glucose (GLU) were significantly decreased in three prescription groups (P<0.05, P<0.01), and the content of high-density lipoprotein cholesterol (HDL-C) was significantly increased (P<0.05, P<0.01). Three prescriptions could decrease the whole blood viscosity and plasma viscosity (P<0.05,P<0.01), and the activated partial thromboplastin time (APTT) could be extended and the content of fibrinogen (FIB) could be decreased (P<0.05, P<0.01). Danshen Drink and Buyang Huanwu Decoction could also significantly prolong the prothrombin time (PT) (P<0.05). The contents of sulfydryl and SOD were significantly increased after the treatment of three prescriptions (P<0.05, P<0.01). In addition, Danshen Drink increased Na+-K+-ATPase activity and decreased the content of malondialdehyde (MDA) (P<0.05, P<0.01). Buyang Huanwu Decoction obviously increased Na+-K+-ATPase activity and the content of sialic acid (P<0.05, P<0.01). Shaofu Zhuyu Decoction significantly increased the content of sialic acid (P<0.01). Conclusion:All three prescriptions show the effects of activating blood and removing blood stasis and hypolipidemic activity. Their common points may be improving the free radical elimination and antioxidant capacity. In addition, three prescriptions also improve the blood stasis state by improving the deformability and aggregation of erythrocytes, so as to activate blood, remove blood stasis and reduce blood lipid.
