目的:采用髌韧带部分缺损联合内侧副韧带离断法建立大鼠膝骨关节炎疾病模型。 方法:将30只SD大鼠随机分为空白组、模型3周组和模型6周组,每组10只。模型3周组和模型6周组采用手术方法造成髌韧带缺损和内侧副韧带离断;空白组只切开皮肤,暴露髌韧带和内侧副韧带,不进行其他手术处理。术后7天开始强迫大鼠定量运动(30 min/d)。分别于造模后3周和6周处死动物,解剖膝关节,暴露髌骨关节面和股骨髁关节面,大体观察关节面形态学改变情况,并采用Pelletier评分表对股骨髁关节面软骨形态进行评分。取膝关节标本,病理切片后行番红O-固绿和阿利新蓝染色,光镜下观察软骨形态结构,改良Mankin’s评分评价其退变程度。 结果:模型3周组和模型6周组大鼠膝关节呈现不同程度的退变,Pelletier和改良Mankin’s评分均高于空白组(P<0.05),且模型6周组评分高于模型3周组(P<0.05)。 结论:采用髌韧带部分缺损联合内侧副韧带离断法可有效建立大鼠膝骨关节炎模型;组织学改变符合骨关节炎软骨退变的病理特征,且手术简单易行、创伤小、感染率低,是较理想的膝骨关节炎造模方法。
Objective:To establish the rat knee osteoarthritis model by partial patellar ligament defect combined with medial collateral ligament transection. Methods30 SD rats were randomly divided into the blank group, 3-week model group and 6-week model group, 10 rats in each group. The rats in the 3-week model group and 6-week model group were performed the operation to induce the partial patellar ligament defect and medial collateral ligament transection. The rats in the blank group were only cut the skin to expose the patellar ligament and medial collateral ligament, without other surgical treatment. One week after surgery, the rats were forced to exercise for 30 minutes per day. 3 weeks and 6 weeks after modeling,the rats were sacrificed respectively,and the knee joint was dissected. The articular surface of the patella and the femoral condyle were exposed to observe the morphological changes,and the cartilage morphology of femoral condyle articular surface was evaluated by Pelletier’s principle. The samples of knee joint were collected,and the pathological sections were stained with safranin O-fast green and Alcian blue to observe the morphological structure of cartilage by light microscopy,and the degree of degeneration was evaluated by modified Mankin’s score. Results:The degeneration of knee joints with different degrees were observed in the 3-week and 6-week model groups,and the Pelletier score and modified Mankin’s score in these two groups were higher than those in the blank group (P<0.05),and the scores in the 6-week model group were higher than those in the 3-week model group (P<0.05). Conclusion:The rat knee osteoarthritis model can be effectively established by the partial patellar ligament defect combined with medial collateral ligament transection. The histological changes are consistent with the pathological features of osteoarthritis cartilage degeneration,and the operation is simple with small traumas and low infection rates,which is an ideal modeling method.
