目的:观察当归补血汤对糖尿病肾病大鼠肾组织ATF6、CHOP、Caspase-3表达的影响,探讨其肾脏保护机制。 方法:60只SD雄性大鼠随机分为对照组和造模组。造模组大鼠采用尾静脉注射链脲佐菌素(STZ)结合高脂高糖饮食建立糖尿病肾病(DN)模型。造模成功的大鼠随机分为模型组、格列齐特组、当归补血汤高剂量组和低剂量组,每组10只。格列齐特组给予格列齐特缓释片2.68 mg·kg-1·d-1,中药高、低剂量组分别给予当归补血汤7.14 mg·kg-1·d-1和1.78 mg·kg-1·d-1,对照组和模型组给予等量0.9% NaCl溶液,连续给药8周。Western blot和PCR分别检测各组大鼠肾组织活化转录因子6(activating transcription factor 6,ATF6)、生长抑制 DNA 损伤基因 153(C/EBP homologous protein,GADD153/CHOP)、天冬氨酸特异性半胱氨酸蛋白酶-3(cysteinyl aspartate specific proteinase-3,Caspase-3)的蛋白及mRNA表达水平。TUNEL染色检测各组大鼠肾组织阳性细胞凋亡率。 结果:与对照组相比,模型组ATF6、CHOP、Caspase-3 mRNA和蛋白表达均升高,TUNEL阳性细胞率增加(P<0.01)。与模型组相比,格列齐特组及当归补血汤高、低剂量组ATF6、CHOP、Caspase-3 mRNA和蛋白表达水平及TUNEL阳性细胞率显著降低(P<0.01)。当归补血汤高剂量组ATF6、CHOP、Caspase-3 mRNA和蛋白表达水平及TUNEL阳性细胞率显著低于格列齐特组和低剂量组(P<0.01)。 结论:当归补血汤能减少糖尿病肾病大鼠肾组织细胞凋亡,其机制可能与下调ATF6、CHOP、Caspase-3表达及抑制内质网应激有关。
Objective:To observe the effects of Danggui Buxue Decoction on the expressions of activating transcription factor 6 (ATF6),C/EBP homologous protein (CHOP) and cysteinyl aspartate specific proteinase-3 (Caspase-3) in the renal tissue of diabetic nephropathy rats,and discuss its mechanism of protecting kidney. Methods60 male SD rats were randomly divided into the control group and modeling group. The model of diabetic nephropathy(DN)was established in the modeling group by streptozotocin (STZ) with intravenous injection in tail combined with the high fat and high glucose diet. After successful modeling,the rats were randomly divided into the model group,gliclazide group,Danggui Buxue Decoction groups with high- and low-dose,10 rats in each group. The gliclazide group was treated with gliclazide sustained-release tablets at a dose of 2.68 mg·kg-1·d-1. The Danggui Buxue Decoction groups with high- and low-dose were treated with Danggui Buxue Decoction at doses of 7.14 mg·kg-1·d-1 and 1.78 mg·kg-1·d-1,respectively. The control group and the model group were treated with equivalent 0.9% NaCl solution. After administration for continuous 8 weeks,the protein and mRNA expression levels of ATF6,CHOP and Caspase-3 were detected by Western blot and PCR. The apoptosis rate of positive cells in renal tissue of rats in each group was detected by TUNEL staining. Results:Compared with the control group,the mRNA and protein expressions of ATF6,CHOP and Caspase-3 were up-regulated in the model group and the rate of TUNEL positive cells was increased (P<0.01). Compared with the model group,the mRNA and protein expressions of ATF6,CHOP and Caspase-3 were down-regulated in the gliclazide group and Danggui Buxue Decoction groups with high- and low-dose,and the rate of TUNEL positive cells was significantly decreased (P<0.01). The mRNA and protein expressions of ATF6,CHOP and Caspase-3 and the rate of TUNEL positive cells in the Danggui Buxue Decoction group with high-dose were significantly lower than those in the gliclazide group or low-dose group (P<0.01). Conclusion:Danggui Buxue Decoction can reduce the renal cell apoptosis in diabetic nephropathy rats. Its mechanism may be related to the down-regulation of ATF6,CHOP and Caspase-3 and the inhibition on endoplasmic reticulum stress.
