Objective:To study the effects of Changweiqing Decoction on DNA double-strand damage and homologous recombination repair factor ATM and γH2AX in nude mice with subcutaneous xenografts of colon cancer . MethodsThe subcutaneous xenografts model was established in the nude mice by human colon cancer cell line(HCT116). The nude mice were divided into the blank group,oxaliplatin(L-OHP) group,Changweiqing group and Changweiqing plus L-OHP group(combination group). The drug treatment groups were treated with L-OHP,Changweiqing Decoction and Changweiqing Decoction plus L-OHP,respectively. The degree of DNA damage was detected by comet assay. The expressions of homologous recombination factor ATM and γH2AX were detected by Western blot and immunohistochemistry. Results:Compared with the blank group,the tailing phenomenon was observed in the Changweiqing group,L-OHP group and combination group,and the tailing phenomenon was more significant in the combination group(P<0.05). Compared with the blank group,the protein expressions of ATM and γH2AX were obviously up-regulated in the L-OHP group and combination group(P<0.05). Compared with the L-OHP group,the protein expressions of ATM and γH2AX were obviously down-regulated in the combination group(P<0.05,P<0.01). Conclusion:The most serious damage of DNA double-strand breaks can be observed in the Changweiqing plus L-OHP group,which suggests that Changweiqing Decoction can enhance the efficacy of L-OHP. Its mechanism is to reduce the homologous recombination repair effects of colon cancer cells on the chemotherapy damage.
