目的:研究“广派”伤科名中医余子贞经验方龙芨消肿膏外敷对兔膝骨关节炎模型滑膜组织病理及相关炎性因子表达的影响。 方法:将60只雄性新西兰大白兔随机分为空白组、模型组、龙芨消肿软膏组(治疗组)和双氯芬酸二乙胺组(对照组),后两组造模成功后分别给予龙芨消肿软膏及双氯芬酸二乙胺外敷2周,进行动物行动评分后处死取材,取膝关节滑膜组织进行组织病理学观察,并用ELISA法测定IL-1、IL-6、LT、TNF-α、MMP-3的表达。 结果:①与模型组比较,治疗组与对照组均能改善木瓜蛋白酶诱导的膝骨关节炎模型活动情况(P<0.05)。②与模型组比较,治疗组及对照组均能降低膝骨关节炎模型动物滑膜组织MMP-3、IL-1表达(P<0.05,P<0.01),且治疗组及对照组两组之间差异无统计学意义(P>0.05)。③组织病理学方面,与模型组比较,治疗组与对照组均能够改善局部滑膜炎情况(P<0.05)。 结论:龙芨消肿软膏能够改善膝骨关节炎动物活动及疼痛情况,可能通过抑制滑膜增生、减少滑液生成,并通过调控局部相关炎性细胞因子表达发挥作用。
Objective:To study the effects of Longji Xiaozhong Ointment with external application on the pathology of synovial tissue and expressions of related inflammatory factors in rabbit model of knee osteoarthritis,which based on the experience of Yu Zizhen,a famous Chinese medical physician of traumatology in Guang style. Methods60 male New Zealand white rabbits were randomly divided into the blank group,model group,Longji Xiaozhong Ointment group(treatment group)and diclofenac diethylamine group(control group). The model was established in the groups except the blank group,and then the treatment group and control group were treated with Longji Xiaozhong Ointment and diclofenac diethylamine by external application respectively. After administration for 2 weeks,the animal behavior were evaluated by modified Lequesne MG scores,and the rabbits were sacrificed. The synovial tissue of knee joint was collected,the histopathological changes were observed and the expressions of IL-1,IL-6,LT,TNF-α and MMP-3 were detected by ELISA. Results:①Compared with the model group,the activities of rabbits with papain-induced knee osteoarthritis were improved in the treatment group and control group(P<0.05). ②Compared with the model group,the expressions of MMP-3 and IL-1 in the synovial tissue of rabbits with knee osteoarthritis were decreased in the treatment group and control group(P<0.05,P<0.01),and there were no statistically significant differences between the treatment group and control group(P>0.05). ③In histopathology,compared with the model group,the local synovitis was improved in the treatment group and control group(P<0.05). Conclusion:Longji Xiaozhong Ointment can improve the activity and pain of rabbits with knee osteoarthritis. It may be associated with the inhibition of synovial hyperplasia,reduction of synovial fluid production and expression regulation of related inflammatory cytokines.
