目的:观察高糖对体外培养小鼠足细胞活力、凋亡的影响以及小檗碱的保护效果。 方法:通过温度条件培养方法诱导永生代小鼠足细胞株分化成熟,分为正常糖对照组(NG,5 mmol/L)、甘露醇高渗对照组(MA,30 mmol/L)、高糖模型组(HG,30 mmol/L)、高糖(30 mmol/L)+小檗碱治疗组(BBR,1.25 μmol/L),干预24 h、48 h、72 h后收集细胞,采用MTT法检测足细胞活力,用流式细胞仪分析细胞凋亡率,并用Q-PCR法检测podocin和nephrin的mRNA表达水平。 结果:NG对照组和MA对照组之间的细胞活力、凋亡率和凋亡相关分子的mRNA表达水平差异无统计学意义(P>0.05);与NG对照组比较,HG模型组足细胞活力和足细胞凋亡率差异有统计学意义(P<0.01),对podocin和nephrin的mRNA表达有明显的抑制作用(P<0.01),并且这种高糖刺激作用可随着作用时间的延长而增强;与HG模型组比较,BBR治疗组足细胞活力和凋亡率差异有统计学意义(P<0.01),podocin和nephrin的mRNA表达水平明显增加(P<0.01)。 结论:高糖环境可诱导小鼠足细胞凋亡,且这种诱导凋亡作用与高渗环境无关;小檗碱对高糖诱导的足细胞凋亡有保护作用。
Objective:To observe the influence of high glucose on the vitality and apoptosis of mouse podocytes cultured in vitro and the protective effects of berberine. MethodsImmortalized mouse podocytes were induced to differentiation and maturation by temperature conditioned culture. The cells were divided into the normal glucose(NG)control group(5 mmol/L),mannitol(MA)hypertonic control group(30 mmol/L),high glucose(HG)model group(30 mmol/L)and berberine(BBR) treatment group(30 mmol/L high glucose + 1.25 μmol/L BBR). After intervention for 24,8 and 72 hours,the cells were collected. The vitality of podocytes was detected by MTT assay,the cell apoptosis rate was analyzed by flow cytometry,and the mRNA expression levels of podocin and nephrin were determined by Q-PCR. Results:There were no statistically significant differences on the cell vitality,apoptosis rate and mRNA expression levels of apoptosis related molecules between the NG control group and MA control group(P>0.05). Compared with the NG control group,the vitality of podocytes was significantly decreased and the apoptosis rate was significantly increased in the HG model group(P<0.01),and the mRNA expressions of podocin and nephrin were obviously inhibited(P<0.01),and the effects of high glucose stimulation could increase over time. Compared with the HG model group,the vitality of podocytes was significantly increased and the apoptosis rate was significantly decreased in the BBR treatment group(P<0.01),and the mRNA expression levels of podocin and nephrin were obviously increased(P<0.01). Conclusion:The high glucose conditions can induce the mouse podocyte apoptosis,and this effect is independent of the hypertonic environment. Berberine shows protective effects on high glucose induced podocyte apoptosis.
