目的:研究苦参碱对大鼠铜绿假单胞菌(PA)感染肺组织Th1/Th2机制的调节。 方法:40只SPF级SD大鼠,随机分为5组,每组8只,分别为正常对照组、铜绿假单胞菌模型组、苦参碱干预组、左氧氟沙星干预组、左氧氟沙星联用苦参碱组。通过大鼠麻醉、支气管插管注入铜绿假单胞菌方法造模,正常对照组用0.9%NaCl溶液代替铜绿假单胞菌。各组采取相应干预措施。观察大鼠肺组织病理变化,检测肺组织白细胞介素4(IL-4)、干扰素γ(IFN-γ)细胞因子含量,转录因子T-bet、Gata-3的变化;用RT-PCR半定量法检测IL-4 mRNA、IFN-γ mRNA以及T-bet mRNA、Gata-3 mRNA的表达。 结果:苦参碱组、左氧氟沙星组及左氧氟沙星联用苦参碱组肺组织IL-4含量及IL-4 mRNA均较铜绿假单胞菌模型组降低,而IFN-γ水平及IFN-γ mRNA较铜绿假单胞菌模型组升高(P<0.05);苦参碱组、左氧氟沙星组及左氧氟沙星联用苦参碱组中肺组织转录因子Gata-3水平和Gata-3 mRNA表达较铜绿假单胞菌模型组降低(P<0.05),而转录因子T-bet水平和T-bet mRNA表达较铜绿假单胞菌模型组升高(P<0.05)。 结论:苦参碱能调节大鼠铜绿假单胞菌肺部转录因子T-bet和Gata-3表达,诱导大鼠机体由Th2型向Th1型分化,促进IFN-γ炎症因子合成、抑制IL-4合成,从而减轻肺部组织感染损伤。
Objective:To study the regulation of matrine on Th1/Th2 mechanism of Pseudomonas aeruginosa(PA)infected lung in rats. Methods40 SD rats of clean grade were randomly divided into the normal control group,PA model group,matrine group,levofloxacin group and combination group,8 rats in each group. After anaesthesia,the model was established by PA injection with bronchial intubation,while the normal control group was treated with normal saline instead of PA. Each group was treated with the corresponding intervention. The pathological changes of lung tissue were observed. The contents of interleukin-4(IL-4) and interferon-γ(IFN-γ) in lung tissue were detected by ELISA,the protein expressions of T-bet and Gata-3 were determined by Western blot,and the mRNA expressions of IL-4,IFN-γ,T-bet and Gata-3 were detected by semi-quantitative RT-PCR. Results:Compared with the PA model group,the content and mRNA expression of IL-4 in lung tissue were decreased in the matrine group,levofloxacin group and combination group,while the content and mRNA expression of IFN-γ were increased(P<0.05). Compared with the PA model group,the protein and mRNA expressions of Gata-3 in lung tissue were decreased in the matrine group,levofloxacin group and combination group(P<0.05),while the protein and mRNA expressions of T-bet were increased(P<0.05). Conclusion:Matrine can regulate the expressions of transcription factor T-bet and Gata-3 in lung of rats with Pseudomonas aeruginosa infection,induce the differentiation from Th2 to Th1,promote IFN-γ synthesis and inhibit IL-4 synthesis,and thus reduce the pathological infection of lung.
