摘要

目的：通过体外药效评价重楼皂苷Ⅰ对肺癌循环肿瘤细胞(circulating tumor cells,CTC)增殖、凋亡的影响。方法：分别用不同浓度的重楼皂苷Ⅰ干预CTC细胞24 h、48 h、72 h后,采用CCK-8法检测细胞增殖情况,采用流式细胞术和激光共聚焦显微镜分析CTC凋亡与周期情况。结果：重楼皂苷Ⅰ能够显著抑制CTC增殖并具有浓度依赖性,诱导CTC细胞核形态发生明显改变,使凋亡细胞的比例显著升高,并将CTC细胞的周期阻滞在G0/G1期。结论：重楼皂苷Ⅰ通过诱导CTC凋亡,抑制细胞从G0/G1期向S期转化,从而发挥抗肿瘤作用。

Abstract

Objective:To evaluate the effects of Polyphyllin I on proliferation and apoptosis of circulating tumor cells (CTC) of lung cancer in vitro. MethodsThe CTC were intervened by Polyphyllin I at different concentrations for 24,8 and 72 hours. The cell proliferation was determined by CCK-8,and the cell apoptosis and cycle were detected by flow cytometry and confocal laser scanning microscopy. Results:Polyphyllin I could significantly inhibit the CTC proliferation in a concentration-dependent manner,and induce the significant morphological changes of CTC nucleus. The proportion of apoptotic cells was significantly increased,and the cells cycle was stagnated in G0/G1 phase. Conclusion:Polyphyllin I can induce CTC apoptosis and inhibit the cells differentiation from G0/G1 to S phase,which plays an anti-tumor effects.
