Objective:To investigate the regulating effect of serum containing Shoutai Pill on proliferation,invasion and migration of human first trimester trophoblastic cells. Methods Human first trimester trophoblastic cells were isolated and cultured in vitro, and then treated with serum containing Shoutai Pill at high-, medium-and low-dose respectively. Blank serum and serum containing dydrogesterone were taken as the control. The proliferation activity, cell cycle distribution, invasion and migration capacity of trophoblastic cells were detected by methyl thiazolyl tetrazolium (MTT) colorimetry,flow cytometry (FCM) and Transwell experiments respectively. Results: Compared with the control group treated with blank serum,after intervention of serum containing Shoutai Pill on the trophoblastic cells,the activity of cell proliferation and the capacity of invasion and migration were obviously enhanced,the cellular proportion in S period was increased and the cellular proportion in G2/M period was decreased(P<0.05). Compared with low-dose group of Shoutai Pill,the activity of cell proliferation and the capacity of invasion and migration were obviously enhanced in the trophoblastic cells treated with Shoutai Pill at medium-and high-dose,and the change of cell cycle distribution was more obvious(P<0.05). Conclusion: Serum containing Shoutai Pill can promote the activity of proliferation and the capacity of invasion and migration of human first trimester trophoblastic cells,and reduce cell apoptosis.
