目的:探讨清热解毒方对小鼠移植性肝癌抗肿瘤作用及趋化因子的影响。 方法:建立小鼠CBRH-7919原位移植性肝癌模型,随机分为中药低、中、高剂量组和对照组,观察清热解毒方的抗肿瘤效应。采用趋化因子芯片筛选出不同剂量组间的差异基因(差异倍数至少为3),继而使用免疫组化及Western blotting方法对差异基因加以验证。 结果:①不同剂量清热解毒方均有不同程度的抗肿瘤作用(与对照组相比P<0.01),组间比较显示高剂量组抑瘤作用更为明显(P<0.01);②趋化因子芯片检测筛选出CXCL1、CXCL2、CXCL3、CXCR2及IL-1β为差异基因,其中中药干预组CXCL1、CXCL2、CXCL3及IL-1β基因表达下调,CXCR2基因上调,均以高剂量组最为明显(P结论:清热解毒方具有显著下调小鼠移植性肝癌模型中趋化因子CXCL1,CXCL2,CXCL3及IL-1β表达,上调趋化因子受体CXCR2的作用,其抗肿瘤作用的机制可能与此有关。
Objective:To investigate the anti-tumor effect of Qingre Jiedu(QRJD) Formula on transplanted hepatocellular carcinoma in mice and the influence on chemokine. MethodsMice models of CBRH-7919 orthotopic hepatocellular carcinoma were established,and the mice were randomly divided into the control group and Chinese medicine groups with low-,mid- and high-dose. The anti-tumor effect of QRJD Formula was observed. The differential genes (with at least 3 folds) among different dose groups were screened by the chemokine chip,and then the differential genes were verified by immunohistochemistry and Western blotting. Results:① QRJD Formula with different doses all showed anti-tumor effect in varying degree(P<0.01 compared with the control group),comparison among groups,the anti-tumor effect of high-dose group was more significant(P<0.01).② CXCL1, CXCL2, CXCL3, CXCR2 and IL-1β were screened as differential gene by the chemokine microarray detection,among,the gene expression of CXCL1, CXCL2, CXCL3 and IL-1β were down-regulated in the Chinese medicine intervention group and the gene expression of CXCR2 was up-regulated,which more significant in the high-dose group (P<0.05).③Above differential gene were verified by immunohistochemistry and Western blotting on the protein levels. Conclusion:QRJD Formula could significantly down-regulate the expression of chemotactic factors CXCL1, CXCL2, CXCL3 and IL-1β and up-regulate the chemotactic factor receptor CXCR2 in mice with transplanted hepatocellular carcinoma,which may be related to its mechanism on anti-tumor.
