Objective:To study the optimum process of ginsenoside Rg,3, prepared by hydrolysis protopanaxadiol-type saponins by glutamic acid. MethodsTaken glutamic acid as catalyst and protopanaxadiol(PPD)-type saponins as raw material,the preparation condition of ginsenoside Rg,3, was optimized by single factor investigation and orthogonal test,and the reaction products were quantitatively analyzed by HPLC. Results:The optimization results of orthogonal test showed that all PPD-type saponins were converted when PPD-type saponins (10 g/L) were hydrolyzed by glutamic acid (10 g/L) at 80℃ for 2 h,the conversion rate of ginsenoside Rg,3, was 65.12%. Conclusion:The method is simple in operation,low in cost with no pollution to environment,and is a feasible method for rapid preparation of 20(S)-ginsenoside Rg,3, and 20(R)-ginsenoside Rg,3,.
