目的:通过观察实验性自身免疫甲状腺炎 (experimental autoimmune thyroiditis,EAT)大鼠甲状腺球蛋白抗体(TGAb)、甲状腺过氧化物酶抗体(TPOAb)、白细胞介素-10(IL-10)及干扰素-γ(IFN-γ)给药前后的变化来探讨青黛对桥本甲状腺炎的疗效及可能机制。 方法:选取SD 雌性大鼠30 只, 适应性喂养1 周,随机取10只为正常对照组,剩余为造模组。造模组给予高浓度碘水喂养,正常对照组给予蒸馏水。第4周起造模组使用完全弗氏佐剂(CFA)的猪甲状腺球蛋白(pTG) 造模,将造模成功后大鼠随机分为模型对照组(10 只)、中药治疗组(10只)。除正常对照组、模型对照组外,中药治疗组给予青黛水煎剂灌胃治疗。各组实验大鼠于治疗第42日后采用酶联免疫吸附法(ELISA) 检测TGAb、TPOAb、INF-γ、IL-10水平。并取大鼠甲状腺组织行病理切片进行光镜下观察。 结果:中药治疗组血清TPOAb、TGAb浓度较模型对照组下降(P<0.01)。与正常对照组比较,所有造模组血清IL-10水平降低(P<0.01),而中药治疗组IL-10的水平比模型对照组升高(P<0.01)。与正常对照组比较,模型对照组和中药治疗组IFN-γ水平显著升高(P<0.01);与模型对照组比较,中药治疗组的IFN-γ水平下降(P<0.01)。 结论:青黛通过调节模型大鼠TGAb、TPOAb、IL-10及IFN-γ水平,从而改善桥本甲状腺炎。
Objective:To investigate the effect of indigo on Hashimoto’s thyroiditis and possible mechanism through comparison of the changes of thyroid globulin antibody (TGAb), thyroid peroxidase antibody (TPOAb), interleukin-10 (IL-10) and interferon-γ(IFN-γ) in experimental autoimmune thyroiditis rats before and after treatment. Methods After adaptive feeding for 1 week,30 SD female rats were randomly divided into the normal control group(10 rats)and modeling group(20 rats). The modeling group was treated with iodine at high concentration, and the normal control group was given distilled water. From 4th week,the modeling group was treated with porcine thyroglobulin (pTG) of complete Freund’s adjuvant (CFA) and the successful modeling rats were randomly divided into the model control group (10 rats) and treatment group of traditional Chinese medicine (10 rats). The treatment group were treated with indigo decoction,with a course of 42 days. The levels of serum TGAb、TPOAb、INF-γ and IL-10 in all groups were tested by ELISA. And the pathological sections of thyroid gland tissue were observed by light microscopy. Results:After treatment, the serum levels of TPOAb and TGAb in the treatment group were decreased compared with the model control group (P<0.01). Compared with the normal control group, the serum IL-10 levels of all modeling groups were decreased (P<0.01), and the levels of IL-10 in the traditional Chinese medicine treatment group were increased compared with the model control group after treatment (P<0.01). Compared with the normal control group, the level of IFN-γ in the model control group and the traditional Chinese medicine treatment group were significantly increased (P<0.01); compared with the model control group, the IFN-γ level of traditional Chinese medicine treatment group was decreased (P<0.01). Conclusion:Indigo regulated the level of TGAb, TPOAb, IL-10 and IFN-γ in model rats,which improved Hashimoto’s thyroiditis.
