目的:观察养心汤对慢性心力衰竭模型大鼠血清脑钠肽(brain natriuretic peptide,BNP)水平和左室射血分数(left ventricular ejection fraction,LVEF)的影响。 方法:选取SD雄性大鼠60只,随机选取10只大鼠作为空白对照组,其余50只大鼠腹腔注射阿霉素(每周4 μg/g),6周后全部大鼠测定心功能,以LVEF≤60%为心力衰竭模型复制成功;将成模大鼠随机分成模型对照组、养心汤组、卡托普利组,每组14只。养心汤组予养心汤浸膏每日2.34 g/kg;卡托普利组予卡托普利每日2.25 μg/g,空白对照组、模型对照组予等体积的蒸馏水。4周后,全部大鼠测定LVEF、血清BNP水平,并采用苏木精伊红(HE)染色观察心肌组织形态。 结果:与空白对照组比较,模型对照组大鼠BNP、LVEF差异有统计学意义(P<0.01);与模型对照组比较,养心汤组和卡托普利组大鼠BNP、LVEF差异有统计学意义(P<0.01);养心汤组与卡托普利组比较, BNP、LVEF差异无统计学意义(P>0.05)。HE染色显示,空白对照组心肌细胞形态、横纵纹、胞质正常,肌纤维排列整齐,胞质均匀,心肌间质未见炎细胞浸润及纤维细胞增生。模型组心肌呈波浪状,间隙增宽、肿胀,炎细胞浸润,部分空泡变性,心肌纤维排列紊乱,间质水肿,甚至出血。养心汤组和卡托普利组可见心肌纤维轻度颗粒样变性,两者间比较无明显差别。 结论:养心汤能降低慢性心力衰竭模型大鼠血清BNP水平,提高左室射血分数,其疗效与卡托普利相当。
Objective:To observe the effect of “Yangxin Decoction” on the serum levels of brain natriuretic peptide (BNP) and left ventricular ejection fraction (LVEF) in rats with chronic heart failure(CHF). Methods60 SD male rats were chose. Among which 10 rats were randomly selected as blank control group,the remaining 50 rats were treated with adriamycin by intraperitoneal injection at the dose of 4 μg/g/week. After 6 weeks the heart function of all rats were measured,and the model of CHF was established successfully with LVEF≤60%. The successful model rats were randomly divided into the model control group, Yangxin decoction group and captopril group, 14 in each group. Yangxin Decoction group was treated with Yangxin decoction extract at the dose of 2.34 g·kg-1·d-1; the captopril group was treated with captopril at the dose of 2.25 μg·g-1·d-1,and the blank control group and model control group were treated with equal volume of distilled water. After 4 weeks,the level of LVEF and serum BNP of all rats were measured,the morphology of myocardial tissue was observed by Hematoxylin-Eosin (HE) staining. Results:For BNP and LVEF,the differences between the blank control and model control group were statistically significance (P<0.01); the differences between the model control and Yangxin Decoction group or captopril group were statistically significance (P<0.01);there were no statistically significant difference between the Yangxin decoction and captopril group (P>0.05). HE staining showed that in the blank control group myocardial cell morphology,ransverse longitudinal lines and cytoplasm were normal,muscle fibers arranged in neat,cytoplasm was homogeneous and no inflammatory cell infiltration and cell proliferation were in the myocardium; while in model group myocardial cells are wavy and the gap widened and swelled,inflammatory cell infiltration,part of the vacuole degeneration,myocardial fiber arrangement disorder, interstitial edema and even bleeding were observed. It can be seen mild myocardial fiber granular degeneration in the Yangxin decoction and captopril group,and there was no obvious difference between two groups. Conclusion:“Yangxin Decoction” could reduce the level of serum BNP and improve LVEF of rats with CHF,the efficacy was comparable with captopril.
