Objective:To use the polytene component (6E,12E)- Tetradecadiene-8,0-diyne-1,3-dioldiacetate (TDEYA) and sesquiterpene component selina-4(14),7(11)-dien-8-one in Atractylodis Japonicae Rhizoma as the control chemicals to establish a TLC qualitative method and a HPLC quantitive method. MethodsTLC method:place samples and control chemicals on pre-coated TLC plate (SIL G-25 UV254); use petroleum ether-ethyl acetate=10:1 as development solvent. The TCL plate was observed under 254 nm UV lamp. HPLC method:ZORBAX SB-C,18, (250 mm×4.6 mm, 5 μm); mobile phase:methanol-1% formic acid solution (67∶33); samples were detected under 254 nm ultraviolet lamp; flow rate:1.0 ml/min; column temperature:30 ℃; injection amount:10 μl. Results:The polytene component TDEYA and sesquiterpene component selina-4(14), 7(11)- dien-8-one were separated well by TLC and HPLC. This method could be used as the qualitative and quantitative methods of evaluating Atractylodis Japonicae Rhizoma. Conclusion: This method is simple in operation, accurate in results and good in reappearance, and can be used as the detection method for evaluating the quality of Atractylodis Japonicae Rhizoma.
