Objective:To observe the effects of electroacupuncture and morphine on hyperalgesia and the activation of spinal astrocyte in rats of bone cancer pain. MethodsSixty Wistar rats were randomly divided into sham operation group, model group, morphine group, electroacupunture group and electroacupuncture with morphine group, 12 rats in each group. From the 15th day after establishing models (inoculating rat mammary gland carcinoma cell Walker 256 into marrow cavity of tibia), the rats were treated accordingly by electroacupuncture and/or morphine once a day for 6 days. Paw withdrawal latency (PWL) was observed after treatments. Expressions of spinal GFAP, IL-1β and NGF were determined by Western blot method and immunofluorescent histochemical staining method. Results:①PWL was longer in the three treatment groups than in the model group (P<0.01), with longer PWL in electroacupuncture and morphine group than in morphine group and electroacupunture group (P<0.01). ②Compared with the model group, expressions of spinal GFAP and NGF in electroacupunture group and electroacupuncture with morphine group were reduced (P<0.01); expressions of spinal IL-1β in the three treatment groups were all reduced (P<0.01). Conclusion:Electroacupuncture with morphine is effective to suppress hyperalgesia in rats of bone cancer pain by inhibiting the activation of spinal astrocyte.
