Objective:To observe the effect of “Xiaotan Sanjie Recipe” on the cell cycle distribution of colorectal cancer cell line,and to study the possible mechanisms. MethodsThe original “Xiaotan Sanjie Recipe” aqueous solution was lyophilized, weighted and dissolved in cell culture and stored. HCT116 cells were treated with different concentrations of “Xiaotan Sanjie Recipe” for 72h,cell cycle distribution was detected by flow cytometry,the expression of proteins(cyclinA2,cyclinB1,CDK2 and CDK1)were detected by Western Blotting. Results:After being treated by “Xiaotan Sanjie Recipe” for 72 h,the proportion of the cells in S phase was increased in 0.47 and 0.94 g/L group, the difference was significant compared with the controlled group(P,<,0.01); the proportion of the cells in G,2, phase were increased in 0.47,0.94 and 1.88 g/L group,the difference was significant compared with the controlled group(P,<,0.01). After being treated by “Xiaotan Sanjie Recipe” for 72 h,the expressions of cyclinA2, CDK2 and CDK1 proteins were significantly decreased in 0.24,0.47,0.94 and 1.88 g/L group,the expression of cyclinB1 was significantly decreased in 0.47,0.94 and 1.88 g/L group,the difference was significant compared with the controlled group(P,<,0.01),the effect was concentration-dependent. Conclusion:“Xiaotan Sanjie Recipe” has the ability to induce the S phase and G,2, phase arrest of HCT116 cell line,its mechanism might be down-regulating the expression of cyclinA2,cyclinB1,CDK2 and CDK1 proteins.
