目的:探讨五味子多糖对微波辐射致生精障碍小鼠的治疗作用及其对生殖激素的影响 方法:50只雄性BALB/C小鼠随机分为正常组、模型组及五味子多糖低、中、高剂量组。除正常组外,每组小鼠每天给予1 450 MHz、50 mW/cm,2,的微波照射1 h,连续12天,制备微波致生精障碍小鼠模型。模型制备后,五味子多糖低、中、高剂量组分别给予五味子多糖100、200、400 mg/kg灌胃,正常组和模型组给予等体积0.9%NaCl溶液灌胃,均每天1次,连续30天。末次灌胃后24 h,取小鼠血清、睾丸和附睾。检测各组小鼠精子密度、活率及畸形率;酶联免疫法测定小鼠血清卵泡刺激素(FSH)、黄体生成素(LH)和睾丸组织匀浆睾酮(T)水平;HE染色观察睾丸形态学变化 结果:与正常组比较,模型组精子密度和精子活率降低、精子畸形率升高(P,<,0. 01),血清FSH和LH水平升高、睾丸组织匀浆中T含量降低 (P,<,0.01)。与模型组比较,五味子多糖各剂量组精子密度、精子活率升高,精子畸形率降低,血清FSH 和LH水平降低,睾丸组织匀浆中T含量升高,差异均有统计学意义(P,<,0.01),且作用效果呈剂量依赖性。组织学观察显示,五味子多糖各剂量组小鼠睾丸组织病理性损害均得到改善 结论:五味子多糖对微波致生精障碍小鼠生精功能有明显的改善作用,其作用机制可能与调节生殖激素水平,恢复下丘脑垂体性腺轴的调控功能有关。
Objective:To explore the efficacy of Schizandra Chinensis polysaccharide (SCP) on dyszoospermia of mice exposed to microwave radiation and its effects on reproductive hormones Methods:Fifty male BALB /C mice were randomly divided into normal control group,model group,low dose SCP group,middle dose SCP group and high dose SCP group. Except normal control group,the dyszoospermia mouse models were established by exposed to microwave of 1450 MHz frequency,power density of 50 mW/cm,2, for 1 h each day for 12 consecutive days. After modeling,SCP (100,0 and 400 mg/kg) was intragastrically administered at corresponding dose to the low, middle and high dose SCP groups.Equal volume of 0.9%NaCl solution was given to rats in the normal control group and the model group by gastric gavage. All the medication was performed once daily for 30 successive days. The blood serum, testis and epididymis were withdrawal 24 h after the last intragastric administration. The levels of follicle stimulating hormone (FSH),luteinizing hormone (LH),and testosterone (T) in the testis homogenate were determined by enzymelinked immunosorbent assay. The sperm count,the motility rate,and the teratospermia rate were compared. The morphology of the testis was observed using HE staining Results:Compared with normal control group,the sperm count and the motility rate decreased,the teratospermia rate increased(P,<,0. 01). The serum levels of FSH and LH increased,the T content in the testis homogenate decreased in the model group,showing statistical difference (P,<,0.01). Compared with the model group,the sperm count and the motility rate increased,the teratospermia rate decreased,the serum levels of FSH and LH decreased,the T content in the testis homogenate increased in all SCP groups,showing statistical difference (P,<,0.01 ). All the indices showed dose-dependent manner in the SCP groups. The histologica Results: showed the pathological injury in the testicular tissue was improved in all SCP groups Conclusion:SCP showed obvious therapeutical effects on dyszoospermia of mice exposed to microwave radiation,and its mechanisms might be correlate with improving reproductive hormone level and recovering the regulation function of hypothalamus-hypophysis-gonad axis.
