目的:探讨丹蛭降糖胶囊(DJC)对糖尿病大鼠肝脏PI3K/AKT信号通路的影响 方法:取雄性SD大鼠55只,除正常对照组外,其余动物给予高脂饲料4周,按35 μg/g给大鼠腹腔注射链脲佐菌素(STZ)溶液1次。造模成功后,大鼠再分为模型组,DJC高、低剂量组及吡格列酮组。DJC高、低剂量组分别给予DJC 1.08 g·kg-1·d-1、5.04 g·kg-1·d-1,吡格列酮组给予吡格列酮10 μg·kg-1·d-1,模型组、正常对照组给予等量0.9%NaCl溶液。模型组、药物组继续给予高脂饲料8周,并连续灌胃8周,留取肝脏,逆转录反应及实时荧光定量PCR检测大鼠肝脏组织PI3K、AKT mRNA表达,取血清测定血糖 结果:与正常对照组比较,模型组大鼠PI3K、AKT mRNA表达明显升高(P<0.01);予DJC后,肝组织PI3K、AKT mRNA表达下降,与模型组比较差异有统计学意义(P<0.01,P<0.05)。大鼠造模后血糖明显升高(P<0.01),DJC灌胃后,高、低两组血糖显著下降(P<0.01),疗效与吡格列酮近似(P>0.05)。血糖与PI3K/AKT相关分析显示,二者呈显著正相关 结论:DJC对糖尿病大鼠肝脏PI3K/AKT信号通路过度表达有显著干预作用,可能与保护肝脏、减轻胰岛素抵抗有关。
Objective:The aim of this study is to investigate the effect of “Danzhi Jiangtang Capsule” on PI3K/AKT signaling pathway in the liver of diabetic rats Methods:55 male SD rats were fed with high calorie diet except the normal control group for 4 weeks. A single intraperitoneal injection of streptozotocin (STZ) of 35mg/kg,when the diabetic rodent model was build, the rats were randomly re-divided into 4 groups:model group, high-does group of DJC, low-does group of DJC and pioglitazone group. High-does group of DJC and low-does group of DJC were treated with 1.08 g/(kg·d) and 0.54 g/(kg·d) repectively, pioglitazone group was treated with 10mg/(kg·d), the blank group and the model group were treated with the same volume of saline .Continuous intragastric administration for 8 weeks,took the liver,detected β-actin, PI3K/AKT in liver tissue of rats by reverse transcription reaction and Quantitative Real-time PCR. Total RNA was extracted from the liver tissue of rats with Trizol kit,determinated the blood glucose and blood lipid Results:Expression of PI3K/AKT in model group increased significantly ,DJC evidently inhibited the excessive expression of PI3K/AKT and RNA. Compared with the normal control group (P<0.05 or P<0.01); the blood glucose of rats were increased significantly (P<0.01) compared with. After being given “Danzhi Jiangtang Capsule ” for 8 weeks,the level of glucose was decreased(P<0.01) ,curative effect was similar to Pioglitazone. The blood glucose and the PI3K/AKT correlation analysis showed a significant positive correlation Conclusion:The DJC has a significant intervention effect on the over expression of PI3K/AKT signaling pathway in the liver of diabetic rats,this may be related to the protection of the liver,reduced vascular endothelial inflammatory reaction.
