目的:研究解郁育胞方对慢性应激大鼠动情周期、血清β-内啡肽(β-EP)及胞饮突等的影响,探讨其对慢性应激大鼠子宫内膜容受性的调节机制 方法:80只SPF级SD大鼠按体质量分层法随机分为5组,即正常组、模型组、解郁育胞方低剂量组(解低组)、解郁育胞方中剂量组(解中组)、解郁育胞方高剂量组(解高组)。除正常组外,其余4组大鼠建立慢性应激模型。于刺激第15日开始,解中组每日给予解郁育胞方溶液灌胃(11.2 μl/g);正常组、模型组灌服等体积纯化水(11.2 μl/g);解低组、解高组分别按解中组折算临床有效剂量的1/2和2倍给药,给药容量5.6 μl/g和22.4 μl/g,每日1次。造模后14日起观察大鼠动情周期。5组大鼠分别于造模第28日16∶00按照雌∶雄=2∶1合笼,经阴道涂片确认妊娠后第4日停药,当日16∶00处死大鼠,检测血清β-EP含量,透射电镜观察子宫内膜上皮细胞胞饮突,统计大鼠的妊娠数和平均着床点数 结果:①一般情况和动情周期:正常组、解中组、解高组相对毛发光洁整齐,活泼好动,眼睛有神,摄食基本正常,体质量上升较迅速。解中组、解高组与模型组比较,动情周期明显缩短(P<0.05),而与正常组差异无统计学意义(P>0.05)。②血清β-EP含量:与正常组比较,模型组、解低组β-EP含量明显升高(P<0.05);与模型组比较,解中组、解高组β-EP含量明显降低(P<0.05);解中组、解高组β-EP含量明显低于解低组(P<0.05)。③透射电镜子宫内膜上皮细胞胞饮突表达:与正常组比较,模型组、解低组评分明显降低(P<0.05),而解高组胞饮突评分与正常组差异无统计学意义(P>0.05);与模型组比较,解中组、解高组的胞饮突评分明显高于模型组(P<0.05);解中组、解高组的胞饮突评分明显高于解低组(P<0.05)。 ④与正常组比较,模型组与解低组的大鼠平均着床点数明显减少(P<0.05),而解中组和解高组平均着床点数无明显差异(P>0.05)。与模型组比较,解中组和解高组平均着床点数明显增高(P<0.05)。解中组、解高组平均着床点数明显高于解低组(P<0.05);解中组和解高组之间的平均着床点数无明显差异(P>0.05) 结论:①解郁育胞方能有效降低慢性应激大鼠血清β-EP值,减轻应激对机体的损伤,增加机体抗应激能力,且呈现一定剂量依赖性。②解郁育胞方高剂量能明显提高慢性应激大鼠子宫内膜上皮细胞的胞饮突成熟度。③解郁育胞方可能通过平衡体内由于紧张应激导致的β-EP分布,提高子宫内膜上皮细胞的胞饮突成熟度,从而改善子宫内膜容受性,提高大鼠的平均着床点数。
Objective:To study the impact of “Jieyu Yubao Prescription” on estrous cycle, β-EP and pinopodes in chronic stress rats and discuss the regulatory mechanism of “Jieyu Yubao Prescription” on endometrial receptivity in chronic stress rats Methods:80 SPF SD rats were randomly divided into 5 groups:normal control group, model group, “Jieyu Yubao Prescription” low-dose group, “Jieyu Yubao Prescription” middle-dose group, “Jieyu Yubao Prescription” high-dose group. Except the normal group, the remaining four groups of rats were established drug chronic stress model. From the 15th day of stimulation, the rats in the “Jieyu Yubao Prescription” middle-dose group were given the solution of “Jieyu Yubao Prescription”, intragastric administration capacity was 11.2 μl/g;normal group and model group were fed with an equal volume of purified water (11.2 μl/g); low-dose group and high-dose group were given 1/2 and 2 times of middle-dose group once a day, the administration capacity was 5.6 μl/g and 22.4 μl/g. The estrous cycle of rats was observed from the fourteenth day after model-building. All the rats in the five groups were made to copulate (female∶male=2∶1) 16∶00 on the 28th day of stimulation. On the 4th day after confirmation of pregnancy by Vaginal smears, the rats were killed at 16∶00 at the day of drug withdrawal. Serum β-EP was tested and endometrial VEGF levels was observed by Transmission Electron Microscope. Pregnancy rate and the average of implantation number were calculated Results:① The general situation and estrous cycle:hair of the rats in the normal group, middle-dose group and high-dose group were clean and neat, active and with bright eyes and normal diet, body weights increased rapidly. Compared with model group, the time of estrous cycle of the rats increased obviously (P<0.05), and there was no significant difference compared with normal group(P>0.05).② The serum β-EP content:Compared with normal group, the serum β-EP of model group and low-dose group was significantly increased (P<0.05); compared with model group, the serum β-EP of middle-dose group and high-dose group was significantly decreased (P<0.05); the serum β-EP of middle-dose group and high-dose group was obviously lower that that in the low-dose group (P<0.05). ③ The expression of endometrial VEGF levels:compared with model group, the score of model group and low-dose group decreased obviously(P<0.05); there was no significant difference in score of high-dose group compared with normal group(P>0.05); the score of middle-dose group and high-dose group obviously higher than the normal group(P<0.05); the score of middle-dose group and high-dose group obviously higher than the low-dose group(P<0.05). ④ Compared with normal group, the average of implantation number of rats in model group and low-dose group decreased significantly (P<0.05), while there was no significant difference in middle-dose group and high-dose group (P>0.05). Compared with normal group, the average of implantation number of rats in middle-dose group and high-dose group increased significantly (P<0.05). The average of implantation number of rats in middle-dose group and high-dose group was higher than that of low-dose group (P<0.05); there was no significant difference in middle-dose group and high-dose group (P>0.05) Conclusion:① “Jieyu Yubao Prescription” can improve or reduce chronic stress serum β-EP valuation effectively, and can reduce stress damage to the body, increase the body’s anti-stress ability, and shows a dose-dependent manner. ② “Jieyu Yubao Prescription” high-dose can significantly increase chronic stress in rats VEGF expression in the endometrium. ③ “Jieyu Yubao Prescription” can increase the expression of VEGF by balancing distribution of β-EP of the body due to the tension caused by the stress, and improve endometrial pinopodes maturity of endometrial epithelial cells, thereby improving the endometrial receptivity.
