目的:观察针刺“头四关穴”对家兔局灶脑缺血再灌注损伤后脑内单胺类神经递质及ATP酶含量的影响,探讨其对神经细胞保护的作用机制 方法:将家兔随机分为假手术组、模型组和针刺组,每组又随机分为1 d、3 d 和7 d 3个时间点。采用大脑中动脉线栓法(MCAO) 建立家兔局灶性脑缺血再灌注模型,在脑缺血2 h后进行再灌注。针刺组家兔在脑缺血6 h后采用电针“头四关穴”进行治疗,每日1次。在再灌注后1 d、3 d 和7 d分别处死取材。采用荧光分光光度法测定脑组织中多巴胺(DA)、去甲肾上腺素(NE)、5-羟色胺(5-HT)的水平;比色法检测缺血脑组织Na,+,-K,+, ATP酶、Ca,2+,ATP酶的含量 结果:与假手术组比较,模型组脑组织中DA、NE和5-HT的水平明显降低,1 d时最为明显,随缺血再灌时间延长,上述指标水平有所提高;Na,+,-K,+, ATP 酶和Ca,2+,ATP 酶含量明显下降,差异有统计学意义(P,<,0.01)。与模型组比较,针刺组各时间点DA、NE和5-HT水平,以及Na,+,-K,+, ATP 酶和Ca,2+, ATP 酶含量均有不同程度的增加(P,<,0.01) 结论:针刺“头四关穴”可有效改善脑缺血再灌注损伤家兔脑组织ATP酶代谢,减少脑缺血区单胺类神经递质的释放,抑制单胺类递质的神经毒性作用,从而发挥抗脑缺血损伤的作用。
Objective:To observe the effect on the content of monoamine neurotransmitter and ATP enzyme by needling “Top siguan” in cerebral ischemia reperfusion rabbit,and to discuss the mechanism of nerve cells protection Methods:rabbits were randomly divided into three groups :sham-operated group, model group and acupuncture group,and each group was divided into post-ischemia 1d,3d and 7d different periods subgroups. The focal cerebral ischemia and reperfusion model were established by the method of the middle cerebral artery occlusion(MCAO). They were performed by ischemia 2 h and then used for reperfusion. After 6 h,acupuncture group was accepted treatment by electroacupuncture “top Siguan”,once a day. The rabbits in the three groups were respectively executed and got cerebral tissue in 1 d,3 d and 7 d. Then they were determined the level of dopamine (DA),noradrenaline (NE) and 5-hydroxytryptamin (5-HT) by using fluorospectrophotometry,and detected the content of Na,+,-K,+,-ATP and Ca,2+,-ATP by using colorimetry Results: Compared by sham-operated group in the same time,DA,NE and 5-HT were decreased obviously in model group especially in the 1 d group,but DA,NE and 5-HT somewhat increased with cerebral ischemia reperfusion time gone by. Na,+,-K,+,-ATP and Ca,2+,-ATP decreased obviously, the differences was statically significant (P,<,0.01). Compared with the model group in every time, the level of DA,NE and 5-HT, the content of Na,+,-K,+,-ATP and Ca,2+,-ATP were increased in different degree(P,<,0.01) Conclusion:Acupuncture “Top siguan”can improve ATP enzyme metabolism,can reduce monoamine neurotransmitter release and inhibit monoamine neurotransmitter toxicity effect in cerebral ischemia reperfusion rabbit. so needling “Top siguan”can play a role of resistance ischemic injury.
