目的:研究甘麦大枣汤对“郁证”模型大鼠行为学特征的影响 方法:将40只Wistar雄性大鼠随机分为正常组、模型组、甘麦大枣汤组和盐酸氟西汀组,每组10只。除正常组外,其余各组以慢性不可预知性温和应激配合孤养的方法进行“郁证”模型大鼠复制。28天完成造模后,各组开始给予相应的干预措施,其中模型组、甘麦大枣汤组和盐酸氟西汀组继续以造模方法给予相应的刺激。盐酸氟西汀组每克体质量灌服10 μg的氟西汀,甘麦大枣汤组灌服甘麦大枣汤浓缩药液2 ml(每毫升药液含2 g生药),正常组和模型组灌服等体积的蒸馏水;各组均干预28天。观察体质量变化情况,同时运用旷场实验、糖水消耗实验观察大鼠的行为学变化情况 结果:造模后,模型组、甘麦大枣汤组、盐酸氟西汀组较正常组体质量减轻,水平运动距离缩短,直立次数和糖水量消耗量减少(P<0.05)。干预后(即实验的第56天),甘麦大枣汤组较模型组体质量、水平运动距离、糖水消耗量增加(P<0.05),盐酸氟西汀组较模型组体质量、水平运动距离、直立次数和糖水消耗量增加(P<0.05);干预后甘麦大枣汤组与盐酸氟西汀组比较,各指标差异均无统计学意义(P>0.05) 结论:甘麦大枣汤可通过影响模型大鼠的行为学特征,对“郁证”发挥治疗作用。
Objective:This paper is to investigate the anti-depression effect of classic recipe “Ganmai Dazao Decoction” by observing the change of behavior characteristics in depressed rats Methods:40 Wistar male rats were randomly divided into control group,model group,“Ganmai Dazao Decoction” group and fluoxetine group,10 in each group. The models were established by chronic unpredictable stress combined solitary keeping 28 days and were given 28 days of drug intervention after modeling success. They were carried by open field test and sugar consumption test to observe the behavior change of depressed rats. The experimental data was recorded and statistical analysis Results:Compared with normal control group,Ganmai Dazao Decoction group and fluoxetine group,weight,horizontal movement distance, upright time and the consumption of sugar water in model group, Ganmai Dazao Decoction group and fluoxetine group showed a tendency to decrease significantly(P<0.05). After the 56th day of the test, weight, horizontal movement distance and the consumption of sugar water increased in the Ganmai Dazao Decoction group(P<0.05), while the fluoxetine group were increased than the model group(P<0.05). Compare between the Ganmai Dazao Decoction group and the fluoxetine group,no significant difference was found in the above index(P>0.05) Conclusion:“Ganmai Dazao Decoction” can affect behavioral characteristics in rat model of depression and relieve the depressed-like behavior in depressed rats.
