目的:研究中药复方潜阳合剂对主动脉缩窄致高血压模型大鼠血压及心肌组织血管紧张素II(AngⅡ)水平的影响 方法:采用腹主动脉缩窄致高血压模型,Wistar大鼠随机分成假手术组和造模组,将造模成功的大鼠随机分成模型组、培哚普利组和潜阳合剂3组,每组10只。术后4周开始灌胃,各组药物干预8周。分别于干预后第4周和第8周使用尾动脉仪,连续动态测量大鼠左前肢血压。采用酶联免疫吸附(ELISA)法测定大鼠心肌组织的AngⅡ水平 结果:①腹主动脉缩窄造模后4周:与假手术组比较,各组收缩压均显著增高(P<0.05);舒张压有增高趋势,但差异无统计学意义(P>0.05)。②药物干预4周后,与模型组比较,培哚普利组、潜阳合剂组收缩压均明显降低(P<0.001),舒张压差异无统计学意义(P>0.05)。③药物干预8周后,与模型组比较,培哚普利组、潜阳合剂组收缩压均明显降低(P<0.05),舒张压差异无统计学意义(P>0.05)。④药物干预8周后,与假手术组比较,模型组心肌组织AngⅡ水平增加(P<0.001);与模型组比较,培哚普利组、潜阳合剂组AngⅡ水平明显下降(P<0.001) 结论:潜阳合剂具有降低腹主动脉缩窄致高血压大鼠收缩压的作用,其机制可能与降低AngⅡ水平有关。
Objective:This paper is to study antihypertensive effect of “Qianyang Decoction” in rats with hypertension by abdominal aortic banding Methods:Hypertention model was established by abdominal aortic banding,and a sham operation was performed for controls. Wistar rats were randomly divided into sham-operation group and operation group. After the modeling,operation rats were randomly divided into model group,perindopril group,“Qianyang Decoction” group,there were 10 rats in each group. 4 weeks after operation,rats were gavaged for 8 weeks. The blood pressure of rats was determined by measuring left forelimb blood pressure with caudal artery continuous dynamic measurement instrument Results:①4 weeks after the modeling,compared with the sham-operation group,each group’s systolic blood pressure is significantly increased(P<0.001). There was increased trend in diastolic blood pressure,but there was no statistical difference between each group(P>0.05). ②4 weeks after intervention,compared with the model group,the systolic blood pressure of perindopril group and Qianyang Decoction group both were significantly decreased(P<0.001),while the diastolic blood pressure has no statistical difference between each group(P>0.05). ③8 weeks after intervention,compared with the model group,the systolic blood pressure of perindopril group and Qianyang Decoction group both were significantly decreased(P<0.05),while the diastolic blood pressure has no statistical difference between each group(P>0.05). ④8 weeks after intervention,compared with the sham-operation group,the level of AngⅡof model group was significantly decreased(P<0.001). Compared with the model Group,the level of AngⅡof perindopril group and Qianyang Decoction group are both significantly decreased (P<0.001) Conclusion:Qianyang Decoction has antihypertension effect in rats with abdominal aortic banding,especially lower systolic pressure,the possible mechanism is by inhibiting AngII levels.
