目的:探索基于关节力学失稳建立膝骨关节炎动物模型的方法 方法:将24只新西兰大白兔随机分为正常组、模型4周组和模型8周组。模型组动物采用手术切断内侧副韧带并切除部分髌韧带的方法造模;正常组仅切开皮肤,显露上述结构而不予特殊处理。各组分别于造模后4周、8周时处死动物,取膝关节标本,病理切片后行HE染色,镜下观察软骨组织形态结构,并应用Mankin评分标准评价关节软骨退变情况;取血清样本,检测IL-1β、TNF-α、MMP-13水平 结果:模型4周组和模型8周组的关节软骨标本分别呈现出不同程度的退变表现,两组的Mankin评分均明显高于正常组(P<0.01);与正常组比较,模型4周组、模型8周组动物血清中IL-1β、TNF-α、MMP-13水平均升高,差异具有统计学意义(P<0.01);与模型4周组比较,模型8周组Mankin评分和IL-1β、TNF-α水平明显升高(P<0.01) 结论:采用切断内侧副韧带并切除部分髌韧带的方法可获得较为理想的兔膝骨关节炎模型,且操作简单,动物感染率和死亡率低。
Objective:To explore the method of animal model building of knee osteoarthritis based on joints’ mechanical instability Methods:Twenty-four New Zealand white rabbits were randomly divided into three groups:normal group, 4 weeks model group and 8 weeks model group, with 8 rabbits in each group. The animals of model groups were making osteoarthritis models by operation, with medial collateral ligament (MCL) cutting off and resection of part of patellar ligament. The rabbits in normal group were only operated by skin incision and exposure of ligaments mentioned above. Animals were sacrificed at 4 and 8 weeks after operation in each group, and the serum and knees of each rabbit were taken at the same time. After HE staining, the morphological structure of articular cartilage was observed by microscope and Mankin’s score was made according to the degree of cartilage degeneration. The expressions of IL-1β, TNF-α and MMP-13 in serum were detected by immunohistochemical techniques Results:The degeneration in varying degrees was observed in articular cartilage from 4 weeks model group and 8 weeks model group, whose Mankin’s scores were significantly higher than the samples of normal group (P<0.01). Comparing with the normal group, the level of IL-1β, TNF-αand MMP-13 in serum were higher from the rabbits both of 4 weeks model group and 8 weeks model group statistically (P<0.01) Conclusion:The modeling method by cutting off MCL as well as excision of part of patellar ligament could provide an ideal animal model of knee osteoarthritis, with the advantages of simple operation, low rates of infection and mortality.
