目的:探讨人参、黄芩仿生化提取物(SQBE)对中波紫外线(UVB)照射人永生化角质形成细胞(HaCaT)缺氧诱导因子1α(HIF-1α)表达的影响,阐明SQBE预防UVB辐射所致皮肤癌发生的可能作用机制。 方法:将对数生长期的HaCaT细胞,接种于含有10%胎牛血清DMEM培养基的培养皿中,于37 ℃、5% CO,2,的细胞培养箱内常规培养;分为对照组和SQBE 1组(终浓度为12.5 μg·ml-1的SQBE)、SQBE 2组(终浓度为25.0 μg·ml-1的SQBE)、SQBE 3组(终浓度为50.0 μg·ml-1的SQBE),各组在给药4 h后与对照组同时进行UVB(0、20、40、60 mJ·cm-2)照射;继续培养24 h后,采用MTT法检测细胞生存率,荧光定量PCR法及Western blotting法检测HIF-1α mRNA及HIF-1α蛋白的表达。 结果:与对照组相同照射剂量比较,SQBE 3组的细胞生存率明显提高(P,<,0.05),SQBE 1组和SQBE 2组差异无统计学意义(P,>,0.05);SQBE 3组能够有效降低细胞内HIF-1α mRNA及HIF-1α蛋白的表达。 结论:大剂量人参、黄芩仿生化提取物(SQBE)可通过对细胞内HIF-1α表达的影响,预防UVB辐射导致的皮肤癌发生。
Objective:To discuss the effect of ginseng and scutellaria bionic extract (SQBE) on HIF-1α expression in HaCaT cells irradiated by UVB and to expound the possible mechanism of anti-UVB radiation on skin cancer. MethodsLogarithmic growth phase HaCaT cells were seeded in petri dishes containing 10% fetal bovine serum in DMEM medium, and cultured in 37 ℃, 5% CO,2, incubator; We divided into a control group, SQBE 1 group (12.5 μg·ml-1 SQBE), SQBE 2 group (25.0 μg·ml-1 SQBE), and SQBE 3 group (50.0 μg·ml-1 SQBE), each group was irradiated by UVB (0,0, 40,0 mJ·cm-2) 4 hours after administration. After 24 hours, the cell survival rate was detected by MTT method; expression of HIF-1αmRNA and HIF-1α protein were detected by real-time PCR and Western blotting. Results:Compared with the control group at the same dose, the cell survival rate in the SQBE 3 group was increased statistically (P,<,0.05), and no significant difference was found between SQBE 1 group and SQBE 2 group (P,>,0.05); the expressions of HIF-1αmRNA and HIF-1α protein were decreased markedly in the SQBE3 group. Conclusion:SQBE can prevent the UVB-induced skin cancer by influcing the expression of HIF-1α in cells possibly.
