目的:观察补肾益气方药对衰老大鼠骨骼肌腺苷酸含量及ATP合酶β蛋白表达的影响 方法:将32只雄性SD大鼠随机分为4组:正常组,模型组,中药组,美雄酮组。除正常组外,其余各组每天颈背部皮下注射10%的D-半乳糖(100 mg·kg-1)溶液,制作“拟衰老”大鼠模型。模型大鼠经补肾益气方药或美雄酮干预10周后,采用高效液相色谱(HPLC)-紫外法检测各组大鼠腓肠肌组织ATP、ADP及AMP含量,并计算能荷(EC)值;采用Western blotting方法检测骨骼肌ATP合酶β蛋白表达变化 结果:与正常组比较,模型组的骨骼肌ATP含量及EC值均显著下降,AMP含量显著升高,ATP合酶β蛋白表达显著下降,差异均有统计学意义(P<0.05)。与模型组比较,中药组ADP含量显著下降,EC值显著升高,美雄酮组较模型组ATP含量及EC值显著增高,中药组及美雄酮组的ATP合酶β蛋白表达量显著增高,差异均有统计学意义(P<0.05)。与美雄酮组比较,中药组ADP含量下降,差异有统计学意义(P<0.01) 结论:补肾益气方药可通过提高大鼠骨骼肌ATP含量及EC值,提高骨骼肌ATP合酶β蛋白的表达量,改善骨骼肌能量代谢,延缓机体及骨骼肌衰老进程。
Objective: To observe the effect of Chinese herbs with tonifying kidney and replenishing qi actions on the adenosine content and the protein expression of the ATP synthase β aging rats' skeletal muscles Methods: Thirty-two SD rats were randomized into four groups:normal group, model group, Chinese medicine group and methandienone group. Except the normal group, rats in the other groups were received subcutaneous injection of 10% D-galactose solution (100 mg·kg-1, once a day) for 10 weeks to make the aging models. After 10 weeks of intervention with Chinese herbs and methandienone, the contents of ATP,ADP and AMP in each group were detected by high performance liquid chromatography (HPLC) - UV method to calculate energy charge (EC) value; the protein expression of the ATP synthase beta in skeletal muscles were detected by Western blotting Results: Compared with the nomal group, the ATP content and EC value of skeletal muscles in the model group were decreased, and the AMP content was rised, and the protein expression of the ATP synthase beta was decreased significantly (P<0.05); compared with the model group, the ADP content in Chinese medicine group was declined, and the EC value was rised significantly; the ATP content and EC value in the methandienone group was rised significantly than those of the DC group; the protein expression of the ATP synthase beta was rised significantly in the Chinese medicine group and methandienone group (P<0.05). Compared with the methandienone group, the ADP content in the Chinese medicine group was reduced significantly (P<0.01) Conclusion: Chinese herbs with tonifying kidney and replenishing qi actions can improve the protein expression of the ATP synthase β, improve energy metabolism of skeletal muscles and retard the aging process of body and skeletal muscles by enhancing ATP content and EC value.
