目的:观察参附配伍对心肌缺血血瘀证模型大鼠血液流变学和认知能力的影响 方法:SD大鼠60只,随机分为6组,每组10只,分别为空白对照组(空白组)、模型组、参附注射液腹腔注射组(注射组)、参附注射液口服组(口服组)、参附汤高剂量组(高剂量组)、参附汤低剂量组(低剂量组)。除空白组外,其余各组以寒凝加药物干预的方法建立心肌缺血血瘀证动物模型,给予相应药物干预。观察各组动物全血黏度、血细胞参数、血沉等血液流变学指标,以及认知能力的影响 结果:①与空白对照组比较,模型组大鼠心电、血细胞参数及血液黏度检测结果差异均有统计学意义(P<0.05),表明药物加寒凝成功造成血瘀模型。②与模型组比较,各给药组心电ST振幅偏移减少,低、中、高切值显著降低,MCV、RDW-CV、MPV值显著降低,跳台实验的错误次数显著减少,差异均有统计学意义(P<0.05);注射组、口服组的PDW值、血沉显著降低(P<0.05);注射组、口服组和高剂量组的潜伏时间显著增加(P<0.05)。与注射组比较,低剂量组ST振幅、全血黏度、血细胞参数(MCV、RDW-CV、MPV值)、血液沉降率和跳台实验的错误次数差异均有统计学意义(P<0.05)。注射组、口服组各指标比较,无统计学差异(P>0.05),表明其药效相当。③血液流变学和行为学指标的典型相关性分析结果显示,寒凝血瘀动物的血流变性和认知能力存在相关性 结论:参附配伍可改善模型动物血液流变学性状,同时提高血瘀动物的认知能力。
Objective: To observe the effect of Radix Ginseng and Radix Aconiti Lateralis Preparata on hemorheology and cognitive ability in rats with myocardial ischemia of the blood stasis pattern.Methods Sixty SD rats were randomly divided into six groups:blank control group, model group, intraperitoneal “Shenfu Injection” group, oral “Shenfu Injection” group, high dose of “Shenfu Decoction” group, low dose of “Shenfu Decoction” group, with 10 rats in each group.Except for blank group, the rat models with myocardial ischemia were established.Each group was treated with corresponding measurements.The viscosity, blood cell parameters (MCV, RDW-CV, MPV and PDW) and hemorheologic indicators were observed.The cognitive ability in each group was detected.Results ① Compared with the blank group, there were significant differences in the ECG,blood cell parameters, blood viscosity in the model group (P<0.05), which indicated that the blood stasis models were established successfully.② Compared with the model group, the ST amplitude, low, medium and high shearing rates, value of MCV, RDW-CV and MPV and error of step-down test were decreased significantly (P<0.05); the PDW value and blood viscosity in injection group and administration group were decreased (P<0.05); the latent time in the in injection group, administration group and high dose group were increased (P<0.05).there were significant differences in the ST amplitude, blood viscosity, blood cell parameters (MCV, RDW-CV, MPV), sedimentation rate and error of step-down test between the injection group and low dose of “Shenfu Decoction” group (P<0.05).No statistical significance was found in all indicators of the “Shenfu Injection” groups (P>0.05), which indicated that the clinical efficacy of the two groups were similar.③ Correlation analysis on the resultes of the hemorheology and behavior indicators showed that there was correlation between hemorheology and cognitive competence.Conclusion Combination of Radix Ginseng and Radix Aconiti Lateralis Preparata can improve the cognitive competence by improving the property of hemorheology.
