Study on geographical characteristics of Paederia scandens based on metabolomics and multimodal chemical analysis
To systematically identify the geographical characteristics of
Paederia scandens
and explore its chemical material basis, thereby providing a scientific foundation for quality evaluation and premium resource development of this medicinal material.
Methods
2
A total of 27 batches of
Paederia scandens
from different geographical origins were collected. Non-targeted metabolomics
was employed for holistic profiling, followed by targeted analysis using high-performance liquid chromatography (HPLC) for multi-component quantification and chemical fingerprinting. Pattern recognition methods, including hierarchical cluster analysis (HCA) and principal component analysis (PCA), were applied for data interpretation.
Results
2
The metabolic profile of
Paederia scandens
from Sichuan province showed significant differences compared with that from other origins. A total of 47 key differential metabolites were identified. The phenylalanine metabolism pathway was active, while iridoid biosynthesis appeared to be inhibited. HPLC quantitative analysis confirmed that multiple iridoid components were significantly lower in Sichuan samples. The results of chemical fingerprinting and pattern recognition were highly consistent with the above findings.
Conclusions
2
An integrated "targeted-nontargeted" analytical strategy was employed for the first time to reveal the unique chemical characteristics and metabolic regulatory basis of
Paederia scandens
produced in Sichuan. Accordingly, it is proposed that the contents of iridoid components including asperuloside, paederosidic acid, and paederoside be used as indicators for geographical origin tracing, and that peaks 4 and 5 in the fingerprint be incorporated into the quality standard as "origin-specific characteristic peaks". These findings provide a scientific basis for the quality evaluation, geographical origin tracing, and premium resource development of
