目的
2
③应用实时逆转录PCR(RT-qPCR)技术检测各组斑马鱼的
BACE1
、
PS1
、
PS2
、
CDK5
和
GSK3β
10 μmol/L Aβ
1-42
Objective
2
To investigate the effects of isoginkgetin (ISO) on AlCl
3
-induced zebrafish behavioral disorders and Aβ
1-42
-induced PC12 cell injury, through the regulation of APP cleaving enzymes and microtubule-associated protein Tau kinases.
Methods
2
① Molecular docking was used to predict the binding energies of ISO with APP cleaving enzymes, including β‑secretase 1 (BACE1), presenilin 1 (PS1), presenilin 2 (PS2), glycogen synthase kinase 3β (GSK3β) and cyclin-dependent kinase 5 (CDK5). ②Zebrafish at 3 days post-fertilization (3 dpf) were used as the subjects of the study and divided into the following groups: control (Ctrl), model (AlCl
3
), ISO low, medium and high doses (2, 8, 16 μg/mL), and donepezil (DPZ) groups (
n
=30). The zebrafish were exposed to the treatment for 24 hours, and behavioral responses to light/dark stimulation were monitored using a behavioral tracking system. ③Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) was used to measure mRNA levels of
BACE1
,
PS1
,
PS2
,
CDK5
, and
GSK3β
in zebrafish. ④ PC12 cells were treated with 10 μmol/L Aβ
1-42
to induce a injury model. Cells were treated with 2.5 μmol/L ISO for 24 hours. The groups were: Ctrl, Ctrl+ISO, Aβ
1-42
, and Aβ
1-42
+ISO groups. RT-qPCR and Western blot were used to measure the mRNA and protein levels of
BACE1
,
PS1
,
PS2
,
CDK5
,
GSK3β
, and to assess the protein levels of p-Tau (396). ⑤ The effects of ISO and LiCl, either alone or in combination, on the protein expression of GSK3β and p‑Tau (396) in Aβ
1-42
-induced PC12 cells were assessed, using 10 μmol/L lithium chloride (LiCl) as a control.
Results
2
①ISO had relatively low binding energies with all target factors. ②ISO (low, medium, and high doses) and DPZ significantly increased the movement speed of zebrafish in the AlCl
3
group. ③RT-qPCR results showed that ISO significantly reduced the mRNA levels of
BACE1
,
PS1
,
PS2
,
CDK5
, and
GSK3β
(
P
<
0.05). ④In PC12 cells, compared to the Aβ
1-42
group, 2.5 μmol/L ISO significantly reduced the protein and mRNA levels of BACE1, PS1, PS2, CDK5, and GSK3β (
P
<
0.05), and decreased the level of p-Tau (396) (
P
<
0.05). ⑤Western blot results showed that ISO and LiCl, whether administered alone or in combination, significantly inhibited the protein levels of GSK3β and p-Tau (
P
<
0.05), with no significant differences observed among the treatment groups (
P
>
0.05).
Conclusion
2
ISO exerts neuroprotective effects by inhibiting the levels of Aβ cleaving enzymes and Tau phosphokinases.
