      摘要:Lung cancer is most likely to metastasize to the brain. It is important to explore the pathogenesis of brain metastasis of lung cancer for the prevention, treatment and prognosis of distant metastasis of lung cancer. Based on the theory of “lung-brain correlation”, we aim to explain the correlation between the lungs and brain in terms of qi movement, five elements, visceral manifestations, as well as channels and collaterals, explore the pathogenesis of lung cancer brain metastasis and provide a theoretical basis for the treatment of lung cancer brain metastasis from the lungs by considering the physiological and pathological characteristics of the lungs and brain.

      摘要:ObjectiveTo observe the effect of Modified Xuanbai Chengqi Decoction (MXBCQD) on intestinal flora and immunity of mice with mycoplasma pneumoniae pneumonia (MPP).MethodsThirty male BALB/c mice were randomly divided into blank group (n=10), model group (n=10) and MXBCQD group (n=10). Except for the blank group, the other mice were modeled by nasal drip of mycoplasma pneumonia (MP) international standard strain, and the mice in the MXBCQD group were fed with MXBCQD for 3 days. On the 4th day, the lungs and intestines of mice were taken and the pathological changes were observed by hematoxylin-eosin (HE) staining, and the quantity of MP-DNA in lung tissue and the numbers of Clostridium butyricum and rumen cocci in feces were detected by real-time fluorescence quantitative reverse transcriptase polymerase chain reaction (RT-qPCR). The levels of inflammatory factors such as interferon gamma (IFN-γ), transforming growth factor β (TGF-β), interleukin 6 (IL-6), interleukin 10 (IL-10), interleukin 17 (IL-17) in bronchoalveolar lavage fluid (BALF) and secretory immunoglobulin A (sIgA) in feces were detected by enzyme linked immunosorbent assay (ELISA). The protein expression of Toll-like receptor 4 (TLR4) in the lung and intestine was detected by Western blot method.Results①Pathological sections of the lung and intestine showed that the inflammatory infiltration in the model group was the most severe, while that in the MXBCQD group was milder. ②Compared with the model group, the content of MP-DNA in MXBCQD group was decreased significantly (P<0.05). ③Compared with the blank group, the rumen cocci and Clostridium butyricum in the model group and MXBCQD group were decreased significantly (P<0.05), and the numbers of the rumen cocci and Clostridium butyricum in MXBCQD group were increased significantly compared with the model group (P<0.05). ④Compared with the blank group, the sIgA contents in the model group and MXBCQD group were decreased significantly (P<0.05), while compared with the model group, the sIgA content in MXBCQD group was significantly increased (P<0.05). ⑤Compared with the blank group, the levels of IFN-γ, TGF-β, IL-6, IL-10, and IL-17 in BALF of the model group and MXBCQD group were increased significantly (P<0.05). Compared with the model group, the levels of those indicators were decreased significantly in MXBCQD group (P<0.05). ⑥Gastrococcus flavum and Clostridium butyricum were positively correlated with sIgA content (P<0.05). ⑦Compared with the blank group, the expression levels of TLR4 protein in the lung and intestinal tissues of the model group and the MXBCQD group were increased significantly (P<0.05). Compared with the model group, the expression levels of TLR4 protein in lung and intestinal tissues of the MXBCQD group were decreased significantly (P<0.05).ConclusionMXBCQD can down-regulate the expression of TLR4 and reduce the level of MPP inflammatory factors, which has a good anti-inflammatory effect; at the same time, it can effectively repair lung and intestinal tissue injury, increase the number of intestinal rumen cocci and Clostridium butyricum, and promote the level of sIgA in intestinal mucosa.

      摘要:ObjectiveTo observe the intervention effect of Liuwei Dihuang Decoction on oxidative stress-related indices, bone turnover markers and the protein expression of FOXO1 in femoral in ovariectomized (OVX) rats. And to explore whether Liuwei Dihuang Decoction can exert antioxidant effect through FOXO1 and its mechanism.Methods(1) In animal experiment: Female Wistar rats were divided into normal group, sham-operated group, model group, Progynova group, N-acetylcysteine (NAC) group, Liuwei Dihuang Decoction high- and low-dose groups. Reactive oxygen species of bone marrow, serum antioxidant indices including SOD, CAT, GSH-PX, GSH and bone turnover markers including BALP, OCN, CTX-I, and the protein expression of FOXO1 in femoral were detected after 8 weeks of intervention. (2) In cell experiments, the decoction of Liuwei Dihuang was prepared and then the medicated serum of Liuwei Dihuang Decoction was collected, including medicated serum of control group, medicated serum groups with low-, medium-and high-dose. ①MC3T3-E1 cells were divided into normal group, model group, control medicated serum group, low-, medium- and high-dose medicated serum group of Liuwei Dihuang Decoction and NAC group. Protein expression of FOXO1 in each group was detected to verify the results of animal experiment. ②FOXO1 gene expression of MC3T3-E1 cells was silenced. After verified by Western blot, the cells were divided into normal group, model group, NAC group, high-dose medicated serum group of Liuwei Dihuang Decoction, siNC group, siFOXO1 group, and siFOXO1+ high-dose group of Liuwei Dihuang Decoction medicated serum. The proliferation and ROS levels of cells in each group were detected. ③MC3T3-E1 cells were divided into normal group, model group, NAC group, control medicated serum group and high-dose medicated serum group of Liuwei Dihuang Decoction, and the protein expression of p-FOXO1 in each group was detected.Results(1) In animal experiment: Compared with the sham operation group, SOD, CAT, GSH-PX, GSH, BALP and OCN levels were significantly decreased (P<0.05), while CTX-I, ROS levels and FOXO1 protein expression were significantly increased in the model group (P<0.05). Compared with the model group, SOD, CAT, GSH-PX, GSH, BALP and OCN levels in high-dose group of Liuwei Dihuang Decoction were significantly increased (P<0.05), while CTX-I, ROS levels and FOXO1 protein expression were significantly decreased (P<0.05). (2) In cell experiment: ①Compared with the normal group, the protein expression of FOXO1 in MC3T3-E1 cells treated with H2O2 was significantly increased (P<0.05). The protein expression of FOXO1 in each dose group of medicated serum of Liuwei Dihuang Decoction was lower than that in the model group (P<0.05). ②After silencing the FOXO1 gene in MC3T3-E1 cells, the cell proliferation in the siFOXO1 group at 48 h and 72 h was significantly lower than that in the model group and siNC group (P<0.05), and the level of ROS was significantly higher than that in the model group and siNC group (P<0.05). The cell proliferation in the siFOXO1+high-dose medicated serum of Liuwei Dihuang Decoction was significantly higher than that in the model group and siFOXO1 group at each time point (P<0.05), but significantly lower than that in the high-dose group of medicated serum of Liuwei Dihuang Decoction (P<0.05); the level of ROS was significantly lower than that in the model group and siFOXO1 group (P<0.05), but significantly higher than that in the high-dose group of medicated serum of Liuwei Dihuang Decoction (P<0.05). ③p-FOXO1/GAPDH in the model group was significantly lower than that in the normal group (P<0.05), and p-FOXO1/GAPDH in the high-dose group of medicated serum of Liuwei Dihuang Decoction was significantly higher than that in the model group (P<0.05).ConclusionLiuwei Dihuang Decoction can inhibit ROS levels in bone marrow of OVX rats, increase antioxidant levels in serum, promote bone formation and inhibit bone resorption in OVX rats. The mechanism may be partly related to promoting the phosphorylation of FOXO1 in osteoblasts.

      摘要:This paper reviews the origin and treatment difficulties of abdominal diagnosis in traditional Chinese medicine, analyzes common clinical abdominal syndromes and types of fang-syndrome in post-stroke coma, and summarizes the clinical experience with Professor Huang Huang's “fang-disease-person” classical prescription diagnosis and treatment system and abdominal diagnosis for the treatment of post-stroke coma. Two proven cases are provided.

      摘要:This article introduces Professor Qi Cong's clinical experience in improving endometrial receptivity with Bushen Huoxue method (method for tonifying the kidneys and activating blood). It is considered that the key pathogenesis of low endometrial receptivity is kidney deficiency and blood stasis, and dysregulation of the liver and spleen. In clinical practice, regulating menstruation is viewed as the basis for embryo implantation, and tonifying the kidneys and activating blood is the main treatment method, supplemented by dispersing the liver and strengthening the spleen. Professor Qi's self-developed Qishi Gongqiang I Prescription (Qi's No.1 Prescription for cavitas uteri) can improve the endometrial blood supply and endometrial receptivity, and thereby increase the embryo implantation rate. One proven case is provided.

      摘要:Postherpetic neuralgia (PHN) is a type of persistent and intractable nerve pain, which pertains to the category of collateral diseases in traditional Chinese medicine. Its disease location is consistent with that of collateral diseases, as is its course and the pathogenesis of various forms of pain. The key to the pathogenesis of PHN is the damaged but unrecovered collateral. Therefore, we differentiate three stages of PHN and treat respectively based on the collateral theory. The pathogenesis of the initial stage is toxin damaging the collateral, and the self-formulated Qingdu Shuluo Formula is prescribed to clear heat and remove toxin, smooth the collateral and relieve pain. The pathogenesis of the middle stage is blood stasis obstructing the collateral, and the self-formulated Huoxue Tongluo Formula is prescribed to activate blood circulation and relieve blood stasis, dredge the collateral and relieve pain. The pathogenesis of the advanced stage is collateral deficiency without recovery, and the self-formulated Buxu Huluo Formula is prescribed to benefit qi and nourish yin, protect the collateral and relieve pain. Meanwhile, medicinal pairs and channel-ushering medicinals are selected accordingly with the change of pain location and nature. If the topical pain is persistent and refractory, collateral-pricking and blood-letting should be used to remove stasis, generate new blood, activate the collateral and therefore improve the therapeutic effect.

      摘要:This article introduces the experience of Professor Su Li in treating rheumatoid arthritis (RA) from stasis. It is held that the pathogenesis of RA lies in the deficiency of healthy qi in the interior, and that's when pathogenic qi can settle in, resulting in obstruction of qi and blood and thereafter blockage of channels and meridians. The treatment should always focus on the pathogenesis of “stasis”, using the method of activating blood and resolving stasis throughout the whole disease course. One proven case is provided.

      摘要:Puerperal fever was first recorded in the Huangdi Neijing, and until Synopsis of the Golden Chamber, the diagnosis and treatment of puerperal fever had been initially formed.General Treatise on Causes and Manifestations of All Diseases in the Sui Dynasty for the first time subdivided the etiology of puerperal fever into external and internal causes, and most of the later physicians embraced his insights. In the Yuan Dynasty, Zhu Danxi proposed that "cold and bitter medicines and sweating medicines should not be used for postpartum illnesses", which had a profound impact on physicians in the Ming and Qing Dynasties. In modern times, with the "Eastward Spread of Western Medicine", various doctors tried to integrate western medicine knowledge into the TCM system while opposing the abuse of warming and tonic medicines.

      摘要:ObjectiveTo investigate the effect and possible mechanism of action of Jianpi Jiedu Formula (JPJDF) on metastatic ability and epithelial mesenchymal transition (EMT) of colorectal cancer.Methods①CCK-8 cell viability assay was used to observe the inhibitory effect of JPJDF at different concentrations on the growth of human colorectal cancer cells. Transwell and scratch healing assay were used to detect the effect of JPJDF on the invasion and migration ability of colorectal cancer cells, respectively. Western blot was used to detect the effect of JPJDF on EMT and the expression of the Hippo signaling pathway-related proteins in cells. ②The effect of JPJDF on lung metastases from colorectal cancer and the Hippo signaling pathway was verified via an in-vivo study with the lung metastases model of colorectal cancer. Hematoxylin-eosin (HE) staining was used to detect lung metastases, and immunohistochemistry was used to detect the expression of YAP protein in lung metastases. ③Transwell, scratch healing assay and Western blot were used to detect the effect of JPJDF on the colorectal cancer cell invasion and metastasis and EMT after the Hippo signaling pathway was intervened with XMU-MP-1.Results①JPJDF inhibited the growth of colorectal cancer cells in a dose- and time-dependent manner. The inhibitory effect on the invasion and migration ability of colorectal cancer cells in JPJDF groups was more significant than that in control group (P<0.05), and JPJDF could inhibit the EMT process of colorectal cancer cells. Further studies showed that JPJDF could promote the activation of the Hippo signaling pathway and reduce YAP protein entry into the nucleus. ②HE staining results of lung metastases in vivo showed that JPJDF could inhibit the volume of lung metastases from colorectal cancer, and immunohistochemistry suggested that JPJDF could reduce the expression of YAP protein in metastases. ③Inhibition of the Hippo signaling pathway with XMU-MP-1 could increase the pools of YAP in the nucleus, promote the invasion and migration ability of colorectal cancer cells and the expression of EMT-related proteins, and boost metastases of colorectal cancer. JPJDF could significantly reduce the invasion and migration ability of colorectal cancer cells and slow down the progression of EMT in the group intervened with XMU-MP-1.ConclusionJPJDF can inhibit the metastatic ability of human colorectal cancer cells, and the mechanism of action is related to the activation of the Hippo signaling pathway.

      摘要:ObjectiveTo observe the effect of Fuxian Qiangxin Recipe on related indexes of myocardial remodeling in rats with chronic heart failure (CHF) induced by adriamycin, and to explore the intervention effect of Fuxian Qiangxin Recipe on heart failure.MethodsSixty SPF male Wistar rats were randomly divided into 6 groups: normal group, model group, captopril group, and Fuxian Qiangxin Recipe low, medium and high dose group, with 10 rats in each group. Except for the normal group, other groups were treated with intraperitoneal injection of adriamycin to replicate chronic heart failure models, and then given corresponding intervention measures. After 6 weeks, the general conditions of the rats in each group were observed. The morphological changes of myocardium were compared by HE staining. The cardiac function of the rats in each group was detected by cardiac color Doppler ultrasound. Enzyme-linked immunosorbent assay (ELISA) was used to detect the content of soluble matrix lysin-2 (sST2) in rat serums, and Western blot was used to detect the protein expression of calcineurin (CaN) in rat myocardial tissues.Results① General condition: Compared with the normal group, the rats in the model group showed symptoms of yang deficiency, such as obvious weight loss, less activity, dull hair, dim nose and mouth, excessive urination and thin stool. The symptoms of each treatment groups were improved compared with those of the model group. ② Pathological observation: The myocardial nucleus of the model group was more stained than that of the normal group, showing local vacuolation, indicating cardiomyocyte injury; compared with the model group, the degree of cardiomyocyte injury in each dose group of Fuxian Qiangxin Recipe was alleviated and improved in varying degrees. ③ Cardiac function: compared with the normal group, the left ventricular ejection fraction (LVEF) decreased, the end-systolic diameter (LVIDs) and end-diastolic diameter (LVIDd) increased significantly in the model group (P<0.05), and the LVEF increased significantly and LVIDs decreased significantly in each group of Fuxian Qiangxin Recipe compared with the model group (P<0.05). ④ Detection of sST2 and CaN: Compared with the normal group, the levels of serum sST2 and myocardial tissue CaN in the model group were significantly higher than those in the normal group, while the levels of sST2 and CaN in each dose group of Fuxian Qiangxin Recipe were lower than those in the model group (P<0.05).ConclusionFuxian Qiangxin Recipe can protect myocardium, improve cardiac function and delay myocardial remodeling in rats with heart failure. The mechanism may be related to the inhibition of serum sST2 and myocardial tissue CaN expression.

      摘要:ObjectiveTo observe the effect of Traditional Chinese medicine compound Yixin Formula(YXF) to improve myocardial ischemia-reperfusion injury(MIRI)in rats and its mechanism.MethodsSixty clean-grade Wistar rats were randomly divided into four groups: sham-operated group, myocardial ischemia-reperfusion group (model group), Yixin Formula+myocardial ischemia-reperfusion group (YXF group), Yixin Formula+myocardial ischemia-reperfusion+SIRT1 inhibitor group (YXF+inhibitor group), 15 rats in each group. After adaptive feeding of rats in each group for 7 d, YXF and YXF + inhibitor groups were given YXF 10 mL/kg by gavage for 7 d, and the remaining groups were given an equal volume of 0.9% sodium chloride solution by gavage. Three days and 20 min before surgery, an inhibitor of SIRT1, sirtinol(2 mg/kg), was injected into the tail vein of rats in the YXF + inhibitor group, and an equal volume of 0.9% sodium chloride solution was injected into the tail vein of rats in the other groups. The sham-operated group was only threaded without ligation, while the left anterior descending branch of the coronary artery was ligated for 30 min in the remaining groups to establish a model of myocardial ischemic injury, and the changes of ECG were observed during operation. After reperfusion, ECG was examined to observe whether the reperfusion model was successfully established, and echocardiography and hemodynamics were used to examine the cardiac function of rats in each group. HE staining was used to observe the histomorphological changes of myocardial tissue. TTC staining was used to determine the area of myocardial infarction. TUNEL method was used to detect the apoptosis rate of myocardial cells. The biochemical method was used to detect the levels of CK and LDH in the serum of rats in each group. ELISA was used to detect the levels of SOD, MDA and H2O2 in the serum of rats in each group. The protein expression of SIRT1, PGC-lα, Bcl-2 and Bax was detected by immunohistochemistry and Western blot.Results① Compared with the sham-operated group, the ST segment of the ECG was significantly elevated in the model group, indicating that the rat myocardial ischemia model was successfully established. Echocardiography and hemodynamic tests showed that LVEF, LVFS, LVSP, +dp/dtmax were decreased and LVDP, -dp/dtmax were increased in the model group (P<0.05). TTC staining showed that the myocardial infarct area was increased in the model group (P<0.05). The biochemical results showed that the serum levels of CK, LDH, MDA, and H2O2 were increased, and the level of SOD was decreased in the model rats (P<0.05). The immunohistochemical and Western blot results showed that the expressions of Bcl-2, SIRT1 and PGC-1α proteins were decreased, and Bax expression was increased in the myocardial tissue of the model rats (P<0.05). ② Compared with the model group, the YXF could reduce the area of myocardial infarction. The levels of LVEF, LVFS, LVSP, +dp/dtmax were increased, the levels of LVDP, -dp/dtmax were decreased, the left ventricular ejection volume was increased, and the cardiac function was improved in the YXF group (P<0.05). The levels of CK, LDH, MDA, H2O2 were decreased and the level of SOD was increased in the YXF group. The expressions of Bcl-2, SIRT1 and PGC-1α in myocardial tissues of the rats were increased, and the expression of Bax was decreased in the YXF group (P<0.05). ③ Compared with the YXF group, the CK, LDH, MDA, and H2O2 levels increased and SOD level decreased in the YXF + inhibitor group (P<0.05). The expressions of Bcl-2, SIRT1, and PGC-1α were decreased and the expression of Bax was increased in the myocardial tissues of the YXF+inhibitor group (P<0.05).ConclusionYXF pretreatment can reduce oxidative stress injury and apoptosis and improve MIRI, and its mechanism of action may be related to the regulation of SIRT1/PGC-1α signaling pathway.

      摘要:ObjectiveTo establish a new method for simultaneous determination of 3-O-acetyl-16α-hydroxytrametenolic acid, dehydropachymic acid and pachymic acid in Poria by quantitative analysis of multi-components with single marker (QAMS).MethodsThe determination was performed on Agilent ZORBAX SB-C18 (4.6 mm×250 mm, 5 μm) column with the mobile phase consisted of acetonitrile - 0.1% phosphoric acid aqueous solution at the flow rate of 0.8 mL/min. The injection volume was 10 μL. The column temperature was set at 30 ℃. The detection wavelength was set at 210 nm. The correction factors of the other two components were calculated by using pachymic acid as the internal reference.ResultsThe correction factors of 3-O-acetyl-16α-hydroxytrametenolic acid and dehydropachymic acid calculated by the internal reference pachymic acid were 0.71 and 1.43, respectively. The measurement data of QAMS were not significantly different from those of the external standard method (relative error<5%).ConclusionThe method is simple and accurate, and can be used for the determination of multi-components in Poria, providing a reference for the quality evaluation of Poria.

      摘要:ObjectiveTo control the internal quality of Shichangpu (Acori Tatarinowii Rhizoma) formula granules, the overall characteristic chromatogram by ultra-performance liquid chromatography (UPLC) and the characteristic chromatogram of water-soluble components by high-performance liquid chromatography (HPLC) were established, and the content of uridine, a nucleic acid component, was determined by HPLC.MethodsThe overall UPLC characteristic chromatogram was determined on a Waters ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 μm) with a gradient elution of methanol and 0.1% phosphoric acid aqueous solution as mobile phases at a flow rate of 0.4 mL/min, detection wavelength of 280 nm, column temperature of 35 ℃, and injection volume of 1 μL. The HPLC characteristic chromatogram of water-soluble components and content determination were performed on a SHIMADZU Shim-pack GIST C18-AQ column (4.6 mm×250 mm, 5 μm) with a gradient elution of methanol and 0.1% phosphoric acid aqueous solution as mobile phases at a flow rate of 1 mL/min, detection wavelength of 260 nm, column temperature of 35 ℃, and injection volume of 10 μL.ResultsThe overall characteristic chromatogram of Acori Tatarinowii Rhizoma formula granules was established with 9 characteristic peaks, among which 4 peaks were identified as asarylaldehyde (peak 3), N-trans-feruloyltyramine (peak 4), β-asarone (peak 6), and α-asarone (peak 9), with similarities of 10 bathes of samples ranging from 0.994 to 1.000. The water solubility characteristic chromatogram Acori Tatarinowii Rhizoma formula granules was established with 8 characteristic peaks, among which 5 component peaks were identified as adenine (peak 1), uridine (peak 2), adenosine (peak 3), guanosine (peak 5), and L-phenylalanine (peak 7), with similarities of 23 batches of samples ranging from 0.931 and 0.972. According to the quantitative analysis, the concentration of uridine had a good linear relationship with the peak area in the linear range of 2.086 to 104.300 mg/L (R2=1.000 0), the mean spiked recovery was 100.16%, the RSD value was 2.02%, and the uridine content of 23 batches of Acori Tatarinowii Rhizoma formula granules ranged from 0.037% to 0.061%, showing the stable quality of the formulation granules.ConclusionThe established characteristic chromatograms and content determination methods are simple, reproducible, and specific, and can be used for the quality control of Acori Tatarinowii Rhizoma formula granules.

      摘要:This paper summarized the evolution of the processing methods and the application of processing accessories of Pinelliae Rhizoma by reviewing the ancient literature on processing since the period of Spring and Autumn and Warring States, and summarized the changes of chemical components and their impact on pharmacological effects before and after processing in the modern processing methods of Pinelliae Rhizoma, concluded the contributions of different chemical components to the pharmacological and pharmacodynamic effects of Pinelliae Rhizoma, and the best technological research of different processed products and so on, which would provide a reference for the modern processing and clinical rational use of Pinelliae Rhizoma.

      摘要:This paper reviews the research progress on the mechanism of acupuncture regulating amyloid-β (Aβ) in Alzheimer's disease (AD). The production, deposition and clearance of Aβ plaques play an important role in the pathological mechanism of AD. By regulating Aβ cleavage, neuroglia, mitochondrial and autophagic systems, Tau protein phosphorylation and neurofibrillary tangles (NFT) formation, acupuncture can reduce Aβ production, deposition and promote Aβ clearance, thus exerting neuroprotective effects.

      摘要:Ferroptosis is a specific form of iron-dependent cell death. More and more studies in recent years have suggested that lymphoma cells are highly sensitive to ferroptosis. Traditional Chinese medicine (TCM) has shown good efficacy in the clinical treatment of lymphoma, and some studies have indicated that some of the action targets of TCM are connected with ferroptosis pathways. Based on the mechanism of ferroptosis and its association with lymphoma, we further explore the targets of TCM intervention in lymphoma in this paper to provide a scientific and theoretical basis for TCM diagnosis and treatment of lymphoma.
