摘要:ObjectiveThe study used a network pharmacology approach and molecular docking technology to identify the pharmacological mechanisms of Compound Tripterygium Wilfordii Painkiller Granules( CTWPGs )treating rheumatoid arthritis (RA).MethodsThe main active ingredients and targets of CTWPGs were obtained by using TCMSP, ETCM and BATMAN. RA-related targets were obtained from OMIM, Drug Bank, Gene Cards and TTD databases. Intersection targets were acquired after drug and disease target mapping. Then the protein-protein interaction network was constructed by String database to analyze the interactions and screened the core genes by topology. Finally, the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis of the core genes were carried out through Database for Annotation, Visualization, and Integrated Discovery. The visual network of “active ingredients-targets-signaling pathways” was constructed by Cytoscape3.6.0 software.ResultsA total of 47 ingredients and 34 core genes were obtained. GO enrichment analysis obtained 357 items (P<0.05), including 299 Biological Processes, 21 Cellular Components and 37 Molecular Functions (P<0.05). And results of KEGG enrichment analysis showed 107 signaling pathways (P<0.05), including TNF signaling pathway, Toll-like receptor signaling pathway, and MAPK signaling pathway. Finally, the results suggested that the key active ingredients of CTWPGs included quercetin, luteolin, and triptolide. And core genes were PTGS2, JUN, AKT1, MAPK1, RELA, MAPK14, MAPK3 and TNF. The molecular mocking showed that luteolin and MAPK3, PTGS2, triptolide and MAPK3, TNF, kaempferol and MAPK14, GA30 and MAPK3, TNF, AKT1, PTGS2, baicalein and MAPK3 had good docking activity.ConclusionNetwork pharmacological studies showed that CTWPGs could play an intervention role in RA through multi-ingredients, multi-targets and multi-pathways, which had certain clinical application value.
摘要:The purpose of this paper is to sort out the academic thought and origin of the method of reinforcing yang and enriching yin in Xu’s pediatrics. It is considered that the TCM etiology and pathogenesis of ventricular premature beats in children may be the imbalance of qi and blood, yin and yang, and the external floating of yang deficiency due to the failure of yin deficiency to control yang. The disease is treated with Xiao’er Fumai Decoction by the method of reinforcing yang and enriching yin, and nourishing the heart and stopping palpitation. And one proven case is presented.

摘要:The purpose of this paper is to summarize and analyze the meaning and clinical significance of professor Xu youjia’s core clinical diagnosis and treatment idea of “harmonizing the spleen and stomach” in pediatrics. He puts forward the academic viewpoint that “the child is the body of deficiency and cold”, explains the necessity of “harmonizing the spleen and stomach”, and holds that when using the method of treating different diseases with the same therapy the pathogenesis evolution and syndrome differentiation should be based on the “spleen and stomach”. Professor Xu youjia’s experience is summarized of treating different diseases with the same therapy by flexible application of the method of “harmonizing the spleen and stomach” in the diagnosis and treatment of common diseases, frequently-occurring diseases and difficult diseases in pediatrics. And two proven cases are presented.

摘要:The article introduces Professor Wang Yaoguang’s experience in the clinical diagnosis and treatment of chronic kidney disease combined with sleep disorders. Professor Wang considers that it is more applicable to treat this disease based on five zang viscera. Different syndromes are divided, including deficiency of liver blood, stagnation of liver qi, fluid retained in heart and lung, deficiency of both heart and spleen, deficiency of kidney yin, deficiency of kidney yang, and deficiency of kidney qi. It is proposed that the treatment based on cause of disease should be combined with syndrome differentiation and treatment, and multiple treatment methods could be applied together. One proven case is presented.

摘要:By explaining the meaning of blood stasis and toxin and summarizing the relationship between blood stasis, toxin and ischemic stroke, this paper discussed the prevention, treatment and recovery of ischemic stroke from the microscopic indicators of blood stasis and toxin. The microscopic indicators of blood stasis and toxin could be used as risk factors and markers for the occurrence, aggravation, and recurrence of ischemic stroke. These microscopic indicators reflected changes in the micro-environment of regional tissue during the occurrence and development of ischemic stroke. What’s more, these microscopic indicators not only could prompt and improve the macroscopic syndrome differentiation of ischemic stroke, but also provided a reference for the choice of ischemic stroke treatment plan.

摘要:This paper summarized professor Wang Yiqin’s characteristics of clinical experience in syndrome differentiation and treatment of asthma. Professor Wang claimed that the mixture of deficiency and excess was the pathogenesis of asthma, and the combination of phlegm and blood stasis was the long-standing root of asthma. The treatment principle of asthma was to give priority to the symptoms and give consideration to the fundamental treatment in the attack period, while give priority to the fundamental treatment and give consideration to the symptoms in the remission period. Professor Wang frequently used ancient classic prescriptions and adapted to clinical symptoms. She advocated that qi should be treated first during the attack of asthma, phlegm and blood stasis should be treated at the same time, and spleen and stomach should be protected from beginning to end. According to the clinical characteristics of phlegm dampness in most children as well as phlegm and blood stasis in most adults during the attack period, she proposed Xiaopingchuan formula and Dapingchuan formula added and subtracted from classic prescriptions to treat them separately, made good use of insect drugs and pair of drugs, and paid attention to children’s inspection, auscultation, and mild medication.

摘要:ObjectiveTo explore the regulatory effect of serum containing traditional Chinese medicine compound with the function of tonifying the kidney and disinhibiting dampness on neutrophilic alkaline phosphatase 6(NALP6) signal transduction pathway in fibroblast-like synoviocytes of gouty arthritis (GA) rats.MethodsThe gouty arthritis model was established by adding sodium urate microcrystalline into the rat fibroblast synovial cell culture medium. The drug containing serum which is most suitable for the survival of rat synovial cells was added into the culture medium for cell experiment. The cells were divided into the blank group (normal synoviocytes), the model group (synoviocytes + sodium urate) and the treatment group (synoviocytes + sodium urate + drug-containing serum) and each group carried on the corresponding intervention. The expression of NALP6 mRNA in synovial cells was detected by RT-PCR, the expression of NALP6, NF-κB, IκB by Western blot, the Caspase-1 activity by Colorimetric Method and IL-1β content in synovial cells of each group by ELISA.ResultsIn the model group, compared with the blank group, the expressions of NALP6 mRNA, NALP6 and NF-κB proteins were up-regulated, while the expression of IκB protein was down-regulated. Caspase-1 activity and IL-1β content were increased(all P<0.05). In the treatment group, compared with the model group, the serum containing TCM compound could significantly inhibit the expressions of NALP6 mRNA, NALP6 and NF-κB proteins, inhibit the activity of Caspase-1, reduce the content of IL-1β, and stimulate the expression of IκB protein in fibroblast synovial cells(all P<0.05).ConclusionBlocking the “NALP6-Caspase-1-IL-1β” inflammatory signal pathway may be one of the most important mechanisms for preventing and treating the gouty arthritis through tonifying the kidney and disinhibiting dampness therapy.

摘要:ObjectiveTo investigate the effect of Dachengqi Decoction on the expression of high mobility group protein B1 (HMGB1) and NOD-like receptor protein 3 (NLRP3) inflammasomes in mice with severe acute pancreatitis (SAP) model.MethodsMale C57BL/6 mice were randomly divided into a control group, a model group, and low, medium and high dose groups of Dachengqi Decoction, with 6 mice in each group. The control group was given intraperitoneal injection of 0.9% NaCl solution, and the rest mice were given intraperitoneal injection of caerulein and lipopolysaccharide to induce SAP model. Dachengqi Decoction was given to the low, medium and high dose groups by gavage at 0 h, 8 h and 16 h after the successful establishment of the model, respectively. After 18 h of modeling, the serum myeloperoxidase (MPO), lactate dehydrogenase (LDH) and trypsin levels were detected by the kit. Serum levels of HMGB1, interleukin-1β (IL-1β) and interleukin-18 (IL-18) were detected by ELISA. After HE staining, the morphological changes of pancreatic tissue were observed under light microscope. Immunohistochemistry and Western blot analysis were used to detect the expression levels of P2X purinoceptor 7 (P2X7), NLRP3, apoptosis-associated speck-like protein (ASC) and aspartate-specific cysteine protease 1 (Caspase-1) in pancreatic tissue.Results①The serum levels of HMGB1, IL-1β, IL-18, trypsin, LDH and MPO in the model group were higher than those in the control group (P<0.05); Compared with the model group, the Dachengqi Decoction low dose group had lower level of serum trypsin (P<0.05), and the medium and high dose groups had lower serum levels of HMGB1, IL-1β, IL-18, trypsin, LDH and MPO (P<0.05). ②There were edema, hemorrhage and inflammatory necrosis in the pancreatic tissue of mice in the model group. Dachengqi Decoction could alleviate the above injuries and protect the pancreatic tissue. ③The Dachengqi Decoction medium and high dose groups could significantly inhibit the expression of upstream P2X7 and its components NLRP3, ASC and Caspase-1 of NLRP3 inflammasomes in SAP mice (P<0.05).ConclusionDachengqi Decoction can effectively inhibit the expression of HMGB1, the activation of NLRP3 inflammasomes and the production of inflammatory factors, thus improving the pancreatic function of SAP model mice.

摘要:ObjectiveTo study the role of marein in mediating SIRT3/SREBP-1 signaling pathway to repair renal fibrosis in diabetic nephropathy (DN).MethodsRats’ glomerular mesangial cells (HBZY-1) were induced by high glucose (HG) and palmitic acid (PA) to establish a DN model in vitro. The cells were divided into normal control group, model group and different dosage groups of marein. Inverted microscope was used to observe the changes in the number and morphology of cells in each group. Western blot was used to detect the expression levels of SIRT3/SREBP-1 pathway protein, fibrosis damage-related proteins transforming growth factor-β1 (TGF-β1), p-Smad2, p-Smad3 and Smad4 in each group. Discovery studio was used to molecularly dock marein with silent information regulator 3 (SIRT3). Immunofluorescence method was used to detect the expression and localization of fibronectin (FN) and Collagen Ⅳ of cells in each group.ResultsMarein inhibited the proliferation of HBZY-1 cells induced by HG and PA, which increased the intercellular and decreased the cell density. Marein down-regulated the expression of SREBP-1, TGF-β1, p-Smad2, p-Smad3 and Smad4 proteins in model cells. Marein up-regulated the expression of active SIRT3, and the molecular docking results showed that marein and SIRT3 protein binded well. Marein inhibited the expression of FN and Collagen Ⅳ protein in cells induced by HG and PA.ConclusionMarein may reduce the kidney inflammation and fibrosis damage induced by HG and PA in HBZY-1 cells by activating SIRT3 protein, mediating the SIRT3/SREBP-1 pathway and down-regulating the TGF-β1/Smads signaling pathway protein.
