Isolation and biological activities study of five alkaloid compounds from Cephalotaxus fortunei
Alkaloids compounds were isolated from the dichloromethane fraction of
Cephalotaxus fortunei
using chromatographic techniques such as silica gel, glucan gel LH-20, and semi-preparative high-performance liquid chromatography. The structures of the compounds were elucidated via one-dimensional/two-dimensional nuclear magnetic resonance (1D/2D NMR), high-resolution electrospray ionization mass spectrometry (HR-ESI-MS), and electronic circular dichroism (ECD) analyses. The cytotoxic effect against HeLa cervical cancer cells and protective activity against H
2
O
2
-induced oxidative damage in Human keratinocytes (HaCaT) were assessed using the Alamar Blue assay.
Results

Five alkaloids compounds Cephalocyclidin A-
N
-oxide (1), (
R
)-Fortunine (2), 4-Hydroxycephalotaxine (3), Cephalocyclidin A (4), and Wilsonine (5) were isolated, of which compounds 1 is a new compound.
In vitro
activity assays showed that alkaloid compounds 1~5 had no significant anti-cervical cancer activity, and the median inhibitory concentration (IC
50
) values for these compounds against HeLa cells were all
>
100 μmol/L. Compounds 1~5 all exhibited significant protective effects against H
2
O
2
-induced oxidative damage in HaCaT (
P
<
0.01), and were able to inhibit the increase of reactive oxygen (ROS) levels caused by H
2
O
2
stimulation, among which the differences of compounds 2, 4 and 5 were statistically significant (
P
<
0.05).
Conclusions

The five alkaloid compounds without side chains isolated from
Cephalotaxus fortunei
had no significant inhibitory effect on cervical cancer cells. However, they demonstrated notable protective activity against H₂O₂-induced oxidative damage in HaCaT. These findings suggest a new direction for the efficacy study of natural products derived from
