Chemical profiling of Huangqi Decoction granules and characterization of absorbed components and metabolites in mice plasma following oral administration based on UHPLC⁃Q⁃Exactive Orbitrap HRMS
Objective,2,To analyze and identify the chemical composition and the main absorbed components in mice after oral administration of Huangqi Decoction granules by UHPLC-Q-Exactive Orbitrap HRMS technique.,Methods,2,Chromatographic separation was performed on an Acquity UPLC BEH C,18, column (100 mm×2.1 mm, 1.7 µm), and 0.1% formic acid aqueous solution-methanol as the mobile phase with gradient elution. The mass spectra were collected using electrospray ionization source ESI,+, and ESI,- ,modes with alternating scans and automatic triggering of the secondary mass spectrometry scan function (Full MS/dd-MS,2,). The chemical composition in Huangqi Decoction granules, absorbed components and metabolites in the serum of mice after oral administration of Huangqi Decoction granules were identified by Xcalibur 3.0 software and compared with reference standards, literatures, and database.,Results,2,A total of 167 chemical compositions were identified from Huangqi Decoction granules, and 60 prototypical components and 43 metabolites were detected in the serum, including liquiritigenin, calycosin, formononetin and astragaloside Ⅳ, etc.,Conclusion,2,This study successfully completed the rapid identification of chemical composition from Huangqi Decoction granules, and clarified main blood components after treated with Huangqi Decoction granules, which can provide partial reference for the analysis of effective substances, study of quality control and explore of action mechanism of Huangqi Decoction granules.
