Simultaneous determination of three triterpenoid acids inPoria by improved QAMS with slope correction
Objective,2,To establish a new method for simultaneous determination of 3-,O,-acetyl-16,α,-hydroxytrametenolic acid, dehydropachymic acid and pachymic acid in Poria by quantitative analysis of multi-components with single marker (QAMS).,Methods,2,The determination was performed on Agilent ZORBAX SB-C,18, (4.6 mm×250 mm, 5 μm) column with the mobile phase consisted of acetonitrile - 0.1% phosphoric acid aqueous solution at the flow rate of 0.8 mL/min. The injection volume was 10 μL. The column temperature was set at 30 ℃. The detection wavelength was set at 210 nm. The correction factors of the other two components were calculated by using pachymic acid as the internal reference.,Results,2,The correction factors of 3-,O,-acetyl-16,α,-hydroxytrametenolic acid and dehydropachymic acid calculated by the internal reference pachymic acid were 0.71 and 1.43, respectively. The measurement data of QAMS were not significantly different from those of the external standard method (relative error,<,5%).,Conclusion,2,The method is simple and accurate, and can be used for the determination of multi-components in Poria, providing a reference for the quality evaluation of Poria.
